摘 要:目的:探讨蝙蝠葛酚性碱(PAMD)对人胰腺癌BxPC-3细胞的增殖抑制作用,以及通过观察PAMD对胰腺癌BxPC-3细胞Hedgehog信号通路中Shh、Ptch1 mRNA和蛋白表达的影响,进一步讨论PAMD抗胰腺癌的作用机制及其作用靶点。方法:对人胰腺癌BxPC-3细胞进行体外培养,以不同终浓度的PAMD培养液进行干预,对照组用正常培养液培养,进行以下实验:(1)利用台盼蓝染色测定各组中BxPC-3细胞生长周期;(2)运用MTT法检测各组BxPC-3细胞增殖情况;(3)利用RT-PCR、Western Blot技术检测各组BxPC-3细胞中Shh、Ptch1 mRNA及蛋白的表达。结果:(1)各给药组与对照组相比,癌细胞生长缓慢;(2)PAMD高、低剂量组及5-FU组细胞的增殖抑制率分别为72.35%、44.62%、56.07%;(3)与对照组相比,Shh、Ptch1 mRNA的表达均下降,差异显著(P<0.01),具有统计学意义。结论:PAMD对人胰腺癌BxPC-3细胞有增殖抑制作用,对Hedgehog信号通路关键位点的表达有一定的抑制作用。由此推断,PAMD对胰腺癌的抗癌作用机制可能与Hedgehog信号通路有关。Objective:The present paper explore the PAMD on the anti-proliferation of human pancreatic cancer BxPC-3 cells,and to research the PAMD on Shh,Ptch1 mRNA and protein expression of Hedgehog signaling pathway in BxPC-3 cells,further to study the antitumor function mechanism and targets of PAMD. Methods:On human pancreatic cancer BxPC-3 cells in vitro,with different final concentration to intervene,then the control group with normal nutrient solution culture.(1)Trpan blue staining method was used to measure the growth cycle of the BxPC-3 pancreatic cancer cells group.(2)Applied MTT to test the proliferative inhibition of BxPC-3 pancreatic cancer cells by PAMD.(3)RT-PCR and Western Blot were used to measure the expression of Shh,Ptch1 mRNA in the pancreatic cancer BxPC-3 cells. Results:(1)Pancreatic cancer cell BxPC-3 were growth in logarithmic growth phase at the fourth day,the cells growed slowly in PAMD and 5-FU group treatment group,when compared with that in the control group;(2)PAMD-high、low dose group and 5-FU on cell proliferation inhibition rate were 72.35%,44.62%,56.07%;(3)The expression of Shh,Ptch1 mRNA and protein were decreased in PAMD groups and 5-FU group which were compared with control group,there were significant differences(P<0.01). Conclusion:PAMD can remarkably restrain the growth of the BxPC-3 pancreatic cancer cells,then on Hedgehog signaling pathway key sites have certain inhibition effects,thus the relate members of hedgehog signal pathway might participate the tumorigenesis of pancreatic cancer.
