摘  要:目的探讨氢气菌群制剂对急性缺氧大鼠的抗氧化效应。方法将健康雄性40只SD大鼠随机分为平原生理盐水(NS)组,高原生理盐水(HS)组,高原氢气菌群制剂低(HL)、中(HM)、高剂量(HH)组(10、20、40mg/kg),每组8只。NS组和HS组大鼠经灌胃给予生理盐水2ml,其余大鼠在模拟海拔5000m等量灌胃给药,每天1次,共给药3d。测定各组大鼠血浆中肿瘤坏死因子α(TNF-α)的含量,肝、肺、心肌组织中丙二醛(MDA)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)的含量。结果与HS组比较,HL组大鼠血浆TNF-α含量明显降低(P<0.01),肺、心肌组织中的MDA含量明显降低(P<0.05),肺组织CAT含量及心肌组织SOD含量明显增加(P<0.01或P<0.05),肝脏、心肌组织的GSH含量明显增加(P<0.05);HM组大鼠肺、心肌组织的MDA含量明显降低(P<0.01或P<0.05),肺组织CAT含量明显增加(P<0.01),肺、心肌组织的SOD含量明显增加(P<0.01),肝、肺和心肌组织的GSH含量明显增加(P<0.01或P<0.05);HH组大鼠心肌组织的MDA含量明显降低(P<0.05),肺组织CAT、SOD含量明显增加(P<0.01),肝、肺和心肌组织的GSH含量明显增加(P<0.01)。结论氢气菌群制剂对缺氧大鼠的心肺损伤有一定保护作用。Objective To explore the antioxidant effect of hydrogen bacterial preparation on acute hypoxia rats. Methods Forty Sprague-Dawley rats were designed to 5 groups(8 each): normoxic saline group(NS), high altitude saline group(HS), high altitude low-dose group(HL), high altitude medium-dose group(HM) and high altitude high-dose group(HH) hydrogen bacterial preparation(10, 20 and 40mg/kg, respectively). Rats in NS group and HS group were given 2ml of normal saline by intragastric administration, and in the rest 3 plateau groups were intragastrically administered with preparations of corresponding dosage at a simulated altitude of 5000 m once a day for 3 days. Contents of tumor necrosis factor alpha(TNF-α) in plasma, malondialdehyde(MDA), catalase(CAT), superoxide dismutase(SOD) and glutathione(GSH) in liver, lung and heart were then measured. Results Compared with HS group, the content of plasma TNF-α in HL group was significantly decreased(P<0.01), the contents of MDA in lung and heart were also significantly decreased(P<0.05), while the contents of CAT in lung and SOD in heart were significantly increased(P<0.01 or P<0.05), and of GSH in liver and heart were also increased significantly(P<0.05). In HM group, the contents of MDA in lung and heart were significantly decreased(P<0.01 or P<0.05), while of CAT in lung increased significantly(P<0.01), and of SOD in lung and heart increased significantly(P<0.01), the contents of GSH in liver, lung and heart increased significantly(P<0.01 or P<0.05). The hypoxic rats in HH group, the content of MDA in heart was significantly decreased(P<0.05), the contents of CAT and SOD in lung increased significantly(P<0.01), and the contents of GSH in liver, lung and heart also increased significantly(P<0.01). Conclusion Hydrogen bacterial preparations have a protective effect on injuries of heart and lung in hypoxic rats.
