摘 要:目的本研究旨在研究c-Kit和Sca-1两种干细胞亚型在小鼠多脏器的年龄分布规律,为后续进行相关组织的干细胞抗损伤研究奠定基础。方法选取C57/BL6雄性成年小鼠(3月龄、6月龄、9月龄),分离其骨髓、肝脏、肾脏、肺及胰腺组织,采用组织块酶解法分选各脏器中直径小于30μm的细胞,用CD45、c-Kit及Sca-1荧光抗体标记细胞,流式细胞仪记录CD45^-c、-Kit^+以及CD45^-Sca^-1^+细胞百分比。结果在小鼠骨髓中,CD45^-c-Kit^+细胞数显著多于CD45^-Sca-1^+(P<0.05);而在肝、肾、肺和胰腺组织中,CD45^-Sca^-1^+的细胞数量远多于CD45^-c-Kit^+干细胞(P <0.05)。除胰腺组织,9月龄CD45^-Sca-1^+的细胞数目较3、6月龄有所升高外,其他组织中干细胞的年龄分布趋势差异不显著(P>0.05)。结论 CD45^-c-Kit^+和CD45^-Sca-1^+两种干细胞亚型在小鼠多脏器中均有存在,尤其是数量上占据优势的CD45^-Sca-1^+干细胞可能在多数器官的发育及修复中发挥重要作用。Objective To investigate the age distribution of c-Kit and Sca-1 positive stem cells in multi-organs which can provide more detailed information for the further researches on anti-damage of stem cells in different tissues.Methods C57/BL6 male mice (3months,6months and 9months)were grouped randomly.The cells less than 30μm were isolated by tissue enzyme hydrolysis from bone marrow,liver,kidney,lung and pancreas.These cells were labeled with CD45,c-Kit and Sca-1 fluorescent antibody.The percentage of CD45^-c -Kit^+and CD45^-Sca-1^+stem cells can be calculated by flow cytometry.Results The results indicated that the amount of CD45^-c-Kit^+stem cells was larger than CD45^-Sca -1 ^+stem cells in bone marrow (P <0.05 );However,in liver,kidney, lung and pancreas,the percentage of CD45-Sca-1^+stem cells was higher than CD45 ^-c -Kit^+cells (P <0.05).In pancreas,the percentage of CD45-Sca -1^+in 9months was higher than that of CD45^-c-Kit^+in 3months and 6months group (P <0.05).In other organs,there was no significant difference of CD45^-c -Kit^+and CD45-Sea -1^+stem cells between different age groups.Conclusion These results suggested that CD45 -c -Kit^+and CD45^-Sca -1^+stem cells located in many main organs of mice.Especially,CD45-Sca-1^+stem cells superior in numbers may play an important role in multi -organs'development and repair.
