摘 要:以河西走廊不同生态型芦苇根际和非根际土壤为样品,采用选择性培养基进行了链霉菌的分离,研究了链霉菌的特性及其与生境的关系。共获得链霉菌355株,其中在2mol L-1Na Cl培养基上分离获得链霉菌70株。16S rRNA基因序列聚类分析表明来源于同一类型土壤中的菌株大致聚在一起。细菌和真菌拮抗性测定表明不同土壤中的链霉菌拮抗性差异显著,沼泽土壤和沙丘土壤中的拮抗性链霉菌多于盐化草甸土壤,根际土壤中拮抗性菌株多于非根际土壤,沙丘土壤中的菌株抗菌活性较弱,盐化土壤中拮抗性菌株种类和数量最少,但其抗菌活性最强。菌株的耐盐性与土壤类型密切相关。这些结果说明土壤的异质性决定着可培养链霉菌的种类和适应特征。本研究获得的细菌和真菌拮抗性链霉菌菌株具有进一步研究的价值。The culturable Streptomyces strains were isolated from the rhizosphere and bulk soils of three ecotype reeds in the Hexi Corridor desert. A total of 355 Streptomyces strains were obtained,of which 70 Streptomyces strains were obtained from the medium containing 2 mol L-1 NaCl. The cluster analysis based on the 16 S rRNA gene sequences showed that the Streptomyces strains from the same soil type were approximately clustered together. The number of antibacterial strains from the swamp reed soil and dune reed soil were more than that from the saline meadow reed soil,and the antibacterial Streptomyces number from the rhizosphere soils was more than that from the bulk soils. The isolates from the dune reed soils had lower antimicrobial activity than the others. The saline meadow reed soils had the least antagonistic isolates but the strains had the highest antimicrobial activity.The analysis of salt tolerance of the isolated strains showed that the strain tolerance to salt is closely related to the soil types. In summary,the results suggest that the soil heterogeneity and environmental characteristics determine the species and adaptation of the culturable Streptomyces strains. The Streptomyces isolates obtained in this study are worth further studying.
