摘 要:目的观察白藜芦醇(Res)对大鼠被动回避记忆及海马中NR2A、NR2B和p-NR2B表达的影响。方法将雌性Wistar大鼠随机分为记忆减退模型组、Res 20、40、80 mg/kg干组、雌二醇组和假手术组。用去卵巢合并D-半乳糖腹腔注射的方法建立记忆减退模型。12周后,用避暗实验检测被动回避记忆,用real-time PCR和Western blot检测NR2A和NR2B基因、总蛋白及磷酸化蛋白的表达。结果模型组和假手术组相比,大鼠进入暗室的潜伏期较短(P <0. 01),错误次数增多(P <0. 01),NR2A和NR2B的基因和蛋白表达降低(P <0. 05),p-NR2B/NR2B比值显著升高(P<0. 05)。Res干预组及ERT组与模型组比较,进入暗室的潜伏期延长(P <0. 01),错误次数减少(P <0. 01),NR2A和NR2B基因的表达显著升高(Res干预组、雌二醇组vs模型组:P <0. 05,P <0. 01); 3个Res组及ERT组NR2A和NR2B蛋白表达明显增加(Res 80mg/kg、其余干预组vs模型组:P <0. 01,P <0. 05;)。3个干预组及雌二醇组p-NR2B/NR2B均有下降(Res 80mg/kg、雌二醇组vs模型组:P <0. 01,P <0. 05)。结论 Res上调NR2A和NR2B基因的表达并且促进其蛋白表达,减少p-NR2B的表达,可能是其改善被动记忆的机制之一。Objective To observe the effect of resveratrol( Res) on passive avoidance memory and the expressions of NR2 A,NR2B and p-NR2 B in the hippocampus in rats. Methods Female Wistar rats were randomly divided into 6 groups according to the body weight: the hypomnesia model group( Model),resveratrol-treated groups( Res 20,40,80 mg/kg),estrogen replacement therapy group( ERT) and the Sham control group( Sham control). The hypomnesia model was induced by ovariectomy combined with intraperitoneal injection of D-galactose. At the end of 12 weeks,memory was measured by step through the test. Gene expressions of NR2 A and NR2 B were detected by real-time PCR. The total protein NR2 A,NR2B and the phosph-NR2 B expressions were measured by Western blot. Results Compared with Sham control group,the latency expression of the Model group was shorter( P < 0. 01),and the number of errors increased( P < 0. 01). The gene and protein expressions of NR2 A and NR2 B in the Model group significantly decreased( P < 0. 05). In addition,the ratio of p-NR2B/NR2 B increased significantly in the Model group( P < 0. 05). Compared with the Model group,the latency expressions of Res treated groups and ERT group significantly decreased( P < 0. 01),and the number of errors reduced( P < 0. 01). The gene expressions of NR2 A and NR2 B increased significantly in Res treated groups and ERT group( Res 20,40,80 mg/kg,ERT vs Model: P < 0. 05,P < 0. 01). As to the protein levels,NR2 A and NR2 B expressions up-regulated significantly in the Res treated groups and ERT group( Res 80 mg/kg,other intervention groups vs Model: P < 0. 01,P < 0. 05),and p-NR2B/NR2 B of the Res treated groups and ERT group decreased significantly( Res 80 mg/kg,ERT vs Model: P < 0. 01,P < 0. 05). Conclusion Res can increase the expressions of NR2 A and NR2 B gene and the protein expressions and reduce the phosphorylation of NR2 B protein,which may be one of the mechanisms for Res improving passive avoidance in rats.
