摘 要:目的观察氯化镧暴露对不同发育阶段仔鼠海马组织磷脂酰肌醇-3激酶(PI3K)表达的影响。方法 Wistar雌鼠32只,随机分成对照组及低、中、高氯化镧组,受孕后以饮水方式对实验组动物进行氯化镧暴露,饮水中氯化镧浓度分别为0%、0. 25%、0. 5%和1. 0%,每组8只。分别于仔鼠出生后14d、28d和42d随机抽取仔鼠,采用尼氏染色的方法分析海马CA1区神经细胞尼氏体的表达;采用real-time PCR和western blot法检测海马组织PI3KmRNA及蛋白的表达;采用Morris水迷宫检测仔鼠学习记忆能力。结果氯化镧暴露可影响仔鼠生长发育,引起体重和脑重的下降,海马CA1区尼氏体表达降低,差异均有统计学意义(P <0. 05)。氯化镧组仔鼠在Morris水迷宫的搜索轨迹逐渐偏离隐蔽平台,穿越平台次数减少,差异有统计学意义(P <0. 001)。氯化镧组仔鼠海马中PI3KmRNA和蛋白的表达降低,且与暴露剂量呈负相关(γPI3KmRNA=0. 848和γPI3K蛋白=0. 945),差异有统计学意义(P <0. 001)。结论氯化镧暴露导致海马PI3K表达下降与仔鼠记忆能力受损密切相关。Objective To observe the effects of lanthanum chloride exposure on the expression of Phosphatidylinositol 3-kinase( PI3K) in hippocampus of offspring rats at different developmental stages. Methods 32 Wistar female rats were randomly divided into control group and low,medium and high La Cl3 exposure groups,and 8 animals per group. The animals were exposed to La Cl3 for 0%,0. 25%,0. 5% and 1. 0% after conception. The brain tissues were randomly selected from the offspring rats on the 14 th,28th and 42 nd day after birth. The expressions of Nissl bodies in the hippocampal CA1 neurons were analyzed by Nissl staining. The levels of PI3 K mRNA and protein in the hippocampus were detected by Real-time PCR and western blot. The learning and memory ability was detected by Morris water maze. Results La Cl3 could affect the growth and development of the offspring rats,and cause the decrease of body weight,brain weight and the expression of Nissl bodies,and the differences were statistically significant( P < 0. 05). The search trajectory of La Cl3 exposure groups in the Morris water maze deviated from the concealed platform,and the number of crossing the platform decreased,and the difference was statistically significant( P < 0. 001). The expression levels of PI3 K mRNA and protein in the hippocampus of La Cl3 groups decreased and were negatively correlated with the exposure doses( γPI3K mRNA = 0. 848 and γPI3K protein = 0. 945),and the differences were statistically significant( P < 0. 001). Conclusion Exposure to La Cl3 lead to decreased expression of PI3 K in the hippocampus,which is associated with impaired memory in offspring rats.
