超声生物显微镜定量检测肥胖小鼠左心室形态及功能变化研究

摘  要:目的应用超声生物显微镜评价肥胖小鼠、正常小鼠左心室形态及功能特征。方法将同月龄组的雄性C57BL/6J小鼠(n=40)平均分为对照组(标准饮食,12周),肥胖小鼠组(高脂饮食,12周)。应用超声生物显微镜依次检测肥胖小鼠组和对照组左心室形态及功能,测量指标包括:室间隔厚度(IVSd),左室后壁厚度(LVPWd),左室舒张末期内径(LVEDD),左室收缩末期内径(LVESD),左室舒张末期容积(LVEDV),左室收缩末期容积(LVESV),左室射血分数(LVEF),每个测量指标取5个心动周期的平均值。结果与月龄匹配的对照组小鼠相比较,肥胖小鼠组较对照组IVSd、LVPWd均增厚,LVIDd、LVEDV较对照组小鼠明显增大(P <0. 05),但LVIDs、LVESV在2组间比较差异均无统计学意义(P> 0. 05)。LVEF、MVE、MVA、E/A在2组间比较差异均无统计学意义(P> 0. 05)。结论研究结果表明,在小鼠模型中,肥胖与左心室内径增大及左室壁一致性增厚相关。LVEF、E/A等是肥胖相关的心肌变化的不敏感指标。Objective To analyze the left ventricular( LV) characteristics in the obese mice and the normal mice through Ultrasound biomicroscopy. Methods Mice( n = 40) were equally divided into control group( fed the normal diet for 12 weeks),obesity group( fed the high-fat diet,12 weeks). Animals were sacrificed after 12 weeks and the LV myocardium were collected. All mice were sequentially evaluated by the Ultrasound biomicroscopy. Thickness of inter-ventricular septum( IVSd),LV thickness of posterior wall( LVPWd),LV end-diastolic dimension( LVEDD),LV end-systolic dimension( LVESD) and LV ejection fraction( LVEF) were derived from UBM by averaging data from five cardiac cycles. Results In comparison with those for age-matched control mice,conventional echocardiography results showed that LV remodeling started from 12 weeks in the obese mice,with significant enlargement of IVSd,LVPWd,LVEDD and there was no significant changes in LVESD. The LVEF,MVE,MVA,E/A were similar among all the groups of animals. Conclusion In a mouse model the obesity correlate with the enlargement and the thickness of LV in the mouse model,LVEF and E/A are the insensitive indexes of the myocardial changes associated with obesity.
