摘　　要：目的探究kisspeptin/kiss1r系统在复发性流产患者中的表达及作用机制。方法选取复发性流产绒毛组织(病例组)、正常绒毛组织(对照组)进行研究,实时荧光定量PCR(RT-qPCR)、Western blot、免疫组织化学法检测kisspeptin、kiss1r表达。体外培养JAR、JEG-3细胞株并转染kiss1r,分空白对照组、阴性转染组和kiss1r转染组,CCK8法检测细胞增殖情况,平板细胞克隆实验检测菌落形成,Transwell系统检测细胞迁移、侵袭能力,Western blot检测基质金属蛋白酶(MMP)-2、MMP-9表达。结果与对照组比较,病例组中kisspeptin、kiss1r表达降低(P<0.05)。与kisspeptin、kiss1r阳性表达者相比,kisspeptin、kiss1r阴性表达者促黄体生成素(LH)升高,催乳激素(PRL)、雌二醇(E2)、孕酮(P)降低(P<0.05)。与空白对照组、阴性转染组比较,kiss1r转染组细胞中kisspeptin、kiss1r蛋白表达升高、增殖率升高,细胞菌落数量、迁移、侵袭数量及MMP-2、MMP-9蛋白表达均升高(P<0.05)。结论 kisspeptin、kiss1r在复发性流产患者绒毛组织中呈低表达,与复发性流产有关。Objective To explore the expression and mechanism of kisspeptin/kiss1 rsystem in patients with recurrent spontaneous abortion.Methods Villus tissues from patients with recurrent abortion(the case group)and normal people(the control group)were studied.Real time quantitative PCR(RT-qPCR),Western blot and immunohistochemistry were used to detect the expression of kisspeptin and kiss1r.JAR,JEG-3 cell lines were cultured in vitro and kiss1r was transfected.They were divided into the control group and the negative transfection group and the kiss1r transfected group CCK8 assay was used to detect cell proliferation.Plate cell clone test was used to detect colony formation.Transwell was used to detect cell migration and invasion.Western blot was used to detect matrix metalloproteinase(MMP)-2 and MMP-9 expression.Results Compared with the control group,the expression of kisspeptin and kiss1r decreased in the case group(P<0.05).Compared with the positive expression of kisspeptin and kiss1r,luteinizing hormone(LH),prolactin(PRL),estradiol(E2)and progesterone(P)decreased in kisspeptin and kiss1r negative expressions(P<0.05).Compared with the control group and the negative transfection group,the expression of kisspeptin and kiss1r protein,proliferation rate,cell colonies,numbers of migration and invasion,expression of MMP-2 and MMP-9 all increased in the kiss1r transfected group(P<0.05).Conclusion The low expression of kisspeptin and kiss1r in villus tissues of patients with recurrent abortion is related to recurrent abortion.
