摘  要:LEAFY (LFY)同源基因是控制植物由营养向生殖生长转变的关键基因,在植物花发育过程中发挥重要作用。LFY基因在被子植物中的低拷贝及丰富的变异使其成为解决物种间系统关系的一个代表性基因。本研究通过基因组PCR扩增,获得了小滨菊LFY同源基因LeFL共5个序列。小滨菊LeFL基因的序列长度均为2 764 bp,与甘菊DFL基因的核苷酸同源性为80.66%,包含3个外显子和2个内含子。5个克隆均包含有1 242 bp的完整的编码框,编码413个氨基酸,与拟南芥LFY、金鱼草FLO和甘菊DFL的同源性分别为58%、66%和93%。各克隆间的序列高度保守,核苷酸同源性达到99.9%,推测小滨菊中LFY基因以单拷贝形式存在。利用氨基酸序列构建的系统进化树结果表明LeFL与菊属植物聚在同一枝上,与传统的分类学结果一致。该研究结果为深入了解小滨菊的成花机制及春黄菊族植物的系统进化提供科学依据。LEAFY(LFY) homologous genes are key genes which control the meristem transition from vegetative to inflorescence identity, and play important roles in flowering development. The low copies and rich genetic variety of LFY gene in angiosperm make it be a representative gene to resolve the relationship between species. In this study, the 5 sequences of Le FL, the homologous gene of LFY in L. linearis, were obtained based on the genomic PCR amplification. The sequence length of Le FL gene in L. linearis was 2 764 bp, and its nucleotide homology with DFL gene in Chrysanthemum lavandulifolium was 80.66%. It had three exons and two introns. The 5 clones all contained the complete coding frame of 1 242 bp, encoding 413 amino acids, and its homology with LFY in Arabidopsis thaliana, FLO in Antirrhinum majus and DFL in C. lavandulifolium was 58%, 66% and 93%, respectively. The sequence of each clone was highly conserved, and the nucleotide homology was 99.9%. It was speculated that LFY gene in L. linearis was in the form of single copy. Phylogenetic tree constructed from amino acid sequences showed that Le FL and Chrysanthemum were clustered on the same branch, consistent with the traditional taxonomic results. The results of this study could provide scientific basis for further research on the mechanism of floral in L. linearis and phyletic evolution of Anthemideae species.
