丹参注射液通过调控NF-κB信号通路对卵巢癌细胞A2780增殖的影响

摘　　要：目的研究丹参注射液通过调控NF-κB信号通路对卵巢癌细胞A2780增殖的影响。方法利用MTT法研究不同浓度丹参注射液对人卵巢癌细胞A2780的生长抑制作用;应用Transwell实验和划痕实验检测肿瘤细胞的侵袭能力和迁移能力,流式细胞术分析丹参注射液对人卵巢癌细胞A2780凋亡的影响,通过Western blot方法,检测p65、MMP-2、MMP-9、VEGF、Bcl-2、Bad和cleaved-caspase-3蛋白表达变化。结果MTT法检测发现丹参注射液处理后的人卵巢癌细胞A2780生长受到显著抑制,且表现出明显的剂量、时效依赖性(P<0.05)。Transwell实验和划痕实验结果表明,丹参注射液能够抑制人卵巢癌细胞A2780侵袭能力和迁移能力(P<0.05),并诱导细胞凋亡。Western blot结果分析显示丹参注射液能够抑制p65、MMP-2、MMP-9、VEGF蛋白表达,下调Bcl-2蛋白,促进Bad和cleaved-caspase-3蛋白表达。结论丹参注射液能够通过抑制p65、MMP-2、MMP-9蛋白表达,阻断NF-κB信号通路,降低A2780细胞的侵袭和迁移能力,抑制VEGF和Bcl-2表达,上调Bad蛋白表达,通过诱导细胞凋亡抑制人卵巢癌细胞A2780增殖。Objective To study the effects of Danshen injection on the proliferation of ovarian cancer cell line A2780 by regulating NF-κB signaling pathway.Methods MTT assay was used to study the growth inhibitory effects of different concentrations of Dansheninjection on human ovarian cancer cell line A2780.Transwell assay and scratch assay were used to detect the invasion ability and migration ability of tumor cells.Flow cytometry was used to analyze the Danshen injection on apoptosis of human ovarian cancer cell line A2780,and western blot method was used to detect changes of expression levels of p65,MMP-2,MMP-9,VEGF,Bcl-2,Bad and cleaved-caspase-3 proteins.Results MTT assay showed that the growth of human ovarian cancer cell line A2780 treated with Salvia miltiorrhiza injection was significantly inhibited,and showed a significant doseand time-dependent(P<0.05).Transwell test and scratch test showed that Salvia miltiorrhiza injection could inhibit the invasion and migration of human ovarian cancer cell line A2780(P<0.05),and induce cell apoptosis.Western blot analysis showed that Salvia miltiorrhiza injection could inhibit the expression of p65,MMP-2,MMP-9 and VEGF,down-regulate Bcl-2 protein and promote the expression of Bad and cleaved-caspase-3protein.Conclusion Danshen injection can inhibit the expression of p65,MMP-2 and MMP-9 proteins,block the NF-κB signaling pathway,reduce the invasion and migration of A2780 cell,inhibit the expression of VEGF and Bcl-2,and up-regulate the expression of Bad protein,and it inhibits proliferation of human ovarian cancer cell line A2780 by inducting apoptosis.
