强肌健力方对重症肌无力大鼠Th17/Treg平衡的影响

摘　　要：【目的】观察强肌健力方对实验性重症肌无力(MG)模型大鼠Th17/Treg平衡的影响。【方法】从70只雌性Lewis大鼠中随机选取7只作为正常组,余下63只给予皮下注射鼠源性AchR-α亚单位97-116多肽片段(R97-116)乳化剂复制MG大鼠模型。最终造模成功的大鼠30只,将其随机分为模型组(N=7),强肌健力方高剂量组(N=8)、中剂量组(N=7)、低剂量组(N=8)。强肌健力方高、中、低剂量组给予强肌健力方(剂量分别为23.4、15.6、7.8 g·kg-1·d-1)灌胃,正常组和模型组给予相同体积生理盐水灌胃,给药4周,期间每周2次观察、记录各组大鼠体质量及肌肉力量Lennon评分变化。给药结束后,采用蛋白免疫印迹(Western blot)法检测各组大鼠脾脏组织维甲酸受体相关孤儿受体γt(RORγt)、叉头蛋白p3(Foxp3)、白细胞介素17A(IL-17A)及转化生长因子β(TGF-β)蛋白表达水平。【结果】与模型组比较,强肌健力方各剂量组能够显著提高MG大鼠体质量(P <0.05)及降低Lennon评分(P <0.01),降低RORγt、IL-17A的表达水平(P <0.01),升高Foxp3、TGF-β的表达水平(P <0.05),且其作用与强肌健力方给药剂量呈正相关。【结论】强肌健力方可能通过影响Th17、Treg细胞转录蛋白及其相关细胞因子的表达,调节二者平衡,从而对MG大鼠产生治疗作用。Objective To observe the effect of Qiangji Jianli Recipe on Th17/Treg balance in myasthenia gravis(MG)rats.Methods From 70 female Lewis rats,7 rats were chosen as the normal group,the left 63 rats were given injection of the emulsifier of rat-derived 97-116 peptide(R97-116)of the acetylcholine receptora(AchRα)-subunit to establish the MG model.Ultimately,30 rats were successfully induced into MG model.The 30 MG rats were randomly divided into model group(N=7),high-dose Qiangji Jianli Recipe group(N=8),middle-dose Qiangji Jianli Recipe group(N=7),and low-dose Qiangji Jianli Recipe group(N=8).The high-,middle-and low-dose Qiangji Jianli Recipe groups were given intragastric administration of Qiangji Jianli Recipe(at the dosage of 23.4,15.6,7.8 g·kg-1·d-1,respectively),and the normal group and model group were given intragastric administration of the same volume of normal saline,the treatment lasting for 4 weeks.During the medication period,the body mass and Lennon scores for muscle strength in each group were observed and recorded twice a week.After treatment,the protein expression levels of retinoic receptor-related orphan receptorγt(RORγt),forkhead box p3(Foxp3),interleukin 17A(IL-17A)and transforming growth factorβ(TGF-β)in rat spleen tissues of various groups were detected by Western blotting method.Results Compared with the model group,the body mass was increased(P<0.05)and Lennon scores were decreased(P<0.01),the expression levels of RORγt and IL-17A were lowered(P<0.01)and the expression levels of Foxp3 and TGF-βwere elevated(P<0.05)in various Qiangji Jianli Recipe groups,and the effects were positively correlated with the dosage.Conclusion Qiangji Jianli Recipe has therapeutic effect on the MG rats through modulating the balance of Th17/Treg lymphocytes by affecting the protein expression of transcription factors and related cytokines of Th17 and Treg cells.
