“益气调血、扶本培元”药线灸对多发脑梗死性痴呆大鼠海马ChAT、AChE活性的影响

摘　　要：目的:探讨"益气调血、扶本培元"壮医药线灸疗法对多发脑梗死性痴呆(MID)大鼠海马胆碱乙酰转移酶(ChAT)、乙酰胆碱酯酶(AChE)活性变化的影响。方法:对100只SD大鼠进行造模,筛选后,随机分为正常组、假手术组、模型组、药线灸组、西药组和非穴组,共六组,每组10只。药线灸组予"益气调血、扶本培元"药线点灸治疗,西药组予多奈哌齐溶液灌胃治疗,非穴组予壮医药线点灸两肋下非穴点治疗,剩下的三组予相同时间的捉抓刺激(非真正意义上的治疗手段),干预4周。用比色法检测大鼠海马组织中的ChAT及AChE活性。结果:模型组与正常组比较,ChAT活性显著降低(P<0.01),AChE活性显著升高(P<0.01);药线灸组能显著提高ChAT活性并降低AChE活性(P<0.01),而西药组只能显著降低AChE活性(P<0.01);药线灸组与西药组比较,AChE活性比较差异无统计学意义(P>0.05);非穴组与模型组比较,ChAT和AChE活性均无统计学意义(P>0.05)。结论:"益气调血、扶本培元"药线灸能改善MID大鼠海马ChAT和AChE活性,改善效果优于西药多奈哌齐,并且具有穴位特异性。Objective:Discuss effect of hippocampal ChAT and AChE activity in multiple infaretion dementa rats treated with "YiQiTiaoXue,FuBenPeiYuan" Moxibustion of Zhuang medicated thread.Methods:After modeling and screening,100 SD rats were randomly divided into the normal group,the sham operation group,the model group,the medicinal thread moxibustion group,the western medicine group and the non acupoint group,with six groups,10 rats in each group.The treatment of medicinal thread moxibustion group was " YiQiTiaoXue,FuBenPeiYuan" drug line point moxibustion,the western medicine group was gavage by donepezil solution,the treatment of non acupoint group was to use medicine line point moxibustion to stimulate the non acupoint point under two ribs,the remaining three groups were stimulated with catching with the same time of treatment(non real means of treatment).The course was four weeks.ChAT and AChE activity in the hippocampus of rats was tested by colorimetric method.Results:The activity of ChAT in the model group was significantly lower than that in the normal group(P<0.01),and the activity of AChE higher significantly(P<0.01);The medicinal thread moxibustion group can significantly increase the activity of ChAT and reduce the activity of AChE(P<0.01),but the western medicine group can only reduce the activity of AChE(P<0.01);There was no statistical difference in the activity of AChE between the western medicine group and the medicinal thread moxibustion group(P>0.05);There was no statistical difference in the activity of ChAT and AChE between the non acupoint group and the model group(P>0.05).Conclusion ": YiQiTiaoXue,FuBenPeiYuan" Moxibustion of Zhuang medicated thread can improve the activity of ChAT and AChE in the hippocampus of MID rats,whose effect is better than donepezil.Moreover,this therapy has acupoint specificity.
