摘  要:杜仲为中国特有抗逆多用途树种,基于多个基因协同提升植物抗旱性的研究基础,克隆杜仲(Eucommia ulmoides Oliv.) 3个抗旱同源关键基因EuDREB2A、EuWRKY19和EuXET的保守区域。与数据库中其他28个物种的同源序列,进行其基因家族结构域、理化性质、编码氨基酸序列结构和系统进化关系分析,结果表明:杜仲的3个同源基因分别含有其基因家族保守结构域cd00018、cd03892和cd02176,且保守结构域和理化性质与其他物种基本一致。EuDREB2A基因编码的蛋白不稳定且有3个氨基酸残基缺失,EuXET等电点最高且有5个位点的氨基酸残基与其他物种不同。系统发育进化分析表明,杜仲EuDREB2A与洋白蜡FpDREB2A、EuWRKY19与小麦TaWRKY19、EuXET与胡杨PeXET具有相似的抗旱机制。本研究为EuDREB/WRKY/XET多基因分子辅助育种提供理论和技术基础。Eucommia ulmoides Oliv. is a Chinese indigenous and multipurpose tree species with stress resistance.Based on the research basis of the enhancement of the plant drought resistance by multiple gene collaboration, the conserved domain of 3 homologous key genes(DREB2A, WRKY19, XET) related to drought resistance in Eucommia ulmoides were cloned. The gene family domain, physicochemical properties, the sequence of encoded amino acid and the phylogenetic relationship of these three gene and the homologous sequences of 28 other species obtained from the database were analyzed. The results showed that the three homologous genes of Eucommia ulmoides harbored the conserved domains of the gene family, namely cd00018, cd03892 and cd02176, respectively,and their conserved domain and physicochemical properties were basically the same as those of other species. The protein encoded by EuDREB2 A gene was unstable due to the absence of 3 amino acid residues. The isoelectric point of EuXET conserved peptide was the highest and the amino acid residues of 5 sites were different from other species. Phylogenetic relationships indicated that EuDREB2 A, EuWRKY19 and EuXET of Eucommia ulmoides had the similar drought resistance mechanism with FpDREB2 A of Fraxinus pennsylvanica, TaWRKY19 of wheat and PeXET of Populus euphratica, respectively. This study could provide theoretical and technical basis for EuDREB2A/EuWRKY19/EuXET polygene molecular-assisted breeding.
