摘  要:目的观察周期性张应力对L6大鼠成肌细胞Caspase-12表达的影响。方法成功构建L6大鼠成肌细胞的体外培养力学刺激模型,采用Western-blot法分析周期性张应力对Caspase-12蛋白表达的影响。应用加载频率为10cycles/min,拉伸变形幅度为15%的力,分别给予成肌细胞1,6,12,24h的力学刺激。同时种板对照组与实验组,对照组不给予力学刺激。结果 Caspase-12在加力1h后,蛋白表达量开始升高,加力6h后蛋白表达量显著升高,加力24h达到峰值,与对照组相比,差异均有显著性(P <0. 05)。结论周期性张应力可引起内质网应激(ERS)并最终导致成肌细胞凋亡。Objective To study the effect of cyclic stress on the expression of Caspase-12 in L6 rat myoblasts. Methods An in vitro culture mechanical stimulation model of L6 rat myoblasts was successfully constructed,and the effect of cyclic stress on the expression of Caspase-12 protein was analyzed by Western-blot method. Mechanical stimulation of myoblasts at 1,6,12,24 h was administered with a force of 10 cycles/min and a stretch deformation amplitude of 15%. Meanwhile,mechanical stimulation was not given to the control group and the experimental group. Results The protein expression level of Caspase-12 protein increased after 1h,and increased significantly after 6h of application of force,reaching the peak value at 24 h,and the difference was significant compared with the control group( P < 0. 05). Conclusion Cyclic stress can cause endoplasmic reticulum stress and eventually cause myoblast apoptosis.
