摘 要:中药活性成分高效筛选技术是化学、生物学和医药学等众多领域的前沿研究问题之一。笔者课题组将G-蛋白偶联受体(GPCRs)识别药物的高特异性和高效液相色谱(HPLC)的高分离能力相结合,提出了受体色谱概念,探索了多种受体固定化方法,建立了系列受体色谱模型,成功将其应用于药物-受体相互作用研究及中药活性成分筛选,为中药受体靶向活性成分高效筛选提供了新方法。The high-efficient screening technology of bioactive compounds in Traditional Chinese Medicine(TCM) is one of the main research issues in many fields,including chemistry,biology and medicine science.This paper defined the new concept of receptor chromatography by combining the high recognizing specificity between G-protein-coupled receptors(GPCRs) and their ligands with the high separation capacity of high performance liquid chromatography(HPLC).Subsequent studies were performed to pursue the methods for receptor immobilization and the establishments of desired receptor chromatographic models.These models were validated by the successful application in drug-receptor interaction analysis and the screening of bioactive compounds from traditional Chinese medicine.Receptor chromatography proved to be a new promising alternative for high-efficient screening receptor-targeting bioactive compounds from TCM.
