香菇多糖联合云芝糖肽对Lewis肺癌小鼠免疫调节与抗肿瘤作用的研究

摘　　要：目的:探讨香菇多糖(Lentinan,LNT)和云芝糖肽(Polysaccharopeptide,PSP)联合使用对免疫抑制小鼠和肺癌小鼠的免疫调节作用和抗肿瘤活性。方法:以Lewis肺癌细胞建立肺癌小鼠模型,随机分为空白组(B1组)、生理盐水组(N1组)、PSP+LNT组(C1组)和阿霉素(Adriamycin,ADM)组(A1组);以多柔比星KM小鼠建立免疫抑制模型,随机分为空白组(B2组)、生理盐水组(N2组)和PSP+LNT组(C2组);2模型中B1组和B2组小鼠不行肺癌细胞和ADM注射,2模型中N1组与N2组、C1与C2组在建模后连续20天分别通过灌胃法注入生理盐水0.2 mL和PSP+LNT的混合溶液0.2 mL,A1组小鼠连续3天腹膜内注射4 mg/(kg·d) ADM。采用自动血细胞分析仪对外周血白细胞(WBC)进行计数;比较各组小鼠胸腺指数、脾脏指数和抑瘤作用;采用流式细胞术测定外周血中CD4^+和CD8^+T淋巴细胞,并计算其比值;用微量样本多指标流式蛋白定量方法 (CBA)和小鼠Th1/Th2/Th17细胞因子CBA分析试剂盒测定小鼠血清中白细胞介素-2 (Interleukin-2,IL-2)、肿瘤坏死因子(Tumor necrosis factor,TNF)、干扰素-γ(Interferon-γ,IFN-γ)、白细胞介素-4 (Interleukin-4,IL-4)、白细胞介素-6 (Interleukin-6,IL-6)、白细胞介素-10 (Interleukin-10,IL-10)和白细胞介素-17A (Interleukin-17A,IL-17A)的含量。结果:肺癌模型中,与B1组比较,N1组外周WBC数量、胸腺指数、脾脏指数、CD4^+/CD8^+T比值、TNF、IFN-γ、IL-2和IL-17A均显著降低(P <0.05),血清中IL-10的含量增高(P <0.05);与N1组比较,C1组外周WBC数量、胸腺指数、脾脏指数、CD4^+/CD8^+T比值、TNF、IFN-γ、IL-2和IL-17A均显著提高(P <0.05),血清中IL-10的含量降低(P <0.05),平均肿瘤重量较低(P <0.05);而A1组外周WBC数量、胸腺指数、脾脏指数、CD4^+/CD8^+T比值、TNF、IFN-γ、IL-2和IL-17A明显降低(P <0.05),2组平均肿瘤重量没有差别。免疫抑制模型中,与B2组比较,N2组外周WBC数量、胸腺指数、脾脏指数、CD4^+/CD8^+T比值�Objective:To explore the immunomodulatory effect and antitumor activities of the combined application ofLentinan(LNT)and Polysaccharopeptide(PSP)in immunosuppressive mice and lung cancer mice.Methods:Made lung cancer models of mice by Lewis lung cancer cell C57,and randomly divided them into the blank group(the group B1),the saline group(the group N1),the PSP+LNT group(the group C1)and the adriamycin(ADM)group(the group A1).Made immunosuppressive models by doxorubicin KM mice,and randomly divided them into the blank group(the group B2),the saline group(the group N2)and the PSP+LNT group(the group C2).In these 2 models,the mice in the group B1 and the group B2 were not injected with lung cancer cells and ADM;the group N1 and the group N2 as well as the group C1 and the group C2 were injected with 0.2 mL of saline and the mixed solution of PSP+LNT by gavage for 20 days after modeling;the mice in the group A1 were given 4 mg/(kg·d)ADM by intrathecal injection for 3 days.Counted peripheral white blood cell count(WBC)by automated blood cell analyzer,compared the thymus indexes,spleen indexes and tumor inhibition in each group,and determined CD4^+and CD8^+T lymphocytes in peripheral blood by flow cytometry and calculated the ratio.Measured the content of interleukin-2(IL-2),tumor necrosis factor(TNF),interferon-γ(IFN-γ),interleukin-4(IL-4),interleukin-6(IL-6),interleukin-10(IL-10)and interleukin-17 A(IL-17 A)in serum of mice by means of micro-sample and multi-index cytometric bead array(CBA)and mouse Th1/Th2/Th17 cytokine CBA assay kit.Results:In the lung cancer models,compared with the group B1,the number of peripheral white blood cell count,thymus index,spleen index,CD4^+/CD8^+T ratio,TNF,IFN-γ,IL-2 and IL-17 A in the group N1 were all significantly decreased(P<0.05),and the serum IL-10 content was increased(P<0.05);compared with the group N1,the peripheral white blood cell count,thymus index,spleen index,CD4^+/CD8^+T ratio,TNF,IFN-γ,IL-2 and IL-17 A in the group C1 were all evidently increased(P<0.05),and the s
