摘 要:生物样品前处理技术是药动学研究过程中最重要、最困难和最繁琐的步骤,选择合适的样品前处理技术是进行药动学研究的关键环节。本文综述了近年来药动学研究中广泛使用的样品前处理技术及其特点,包括蛋白沉淀、液液萃取、液相微萃取、固相萃取、固相微萃取、微透析、分子印迹、柱切换、超临界流体萃取。目前,样品前处理的新方法及新技术不断出现,而且正向着简便化、自动化、超灵敏化以及样品、试剂微量化发展。Technology for biological sample pretreatment is the most important,difficult and tedious procedure in pharmacokinetic research.The selection of appropriate technology of sample pretreatment plays a major part in the pharmacokinetic study.In this paper,the techniques of sample pretreatment widely-used in pharmacokinetic studies in recent years are reviewed with the future perspectives,including protein precipitation,liquid-liquid extraction,liquid phase microextraction,solid phase extraction,solid phase microextraction,microdialysis,molecular imprinting,column switching and critical fluid extraction.At present,the new methods and technologies of sample pretreatment are constantly emerging,which are developing towards simplification,automation,hypersensitization and miniaturization of sample sizes and reagent dosages.
