摘 要:目的探讨血清中miRNA-92a家族基因中的miRNA-92a-1-5p和miRNA-92a-3p在健康人群、糖尿病前期、2型糖尿病以及糖尿病视网膜病变等不同疾病阶段的表达变化。方法收集糖尿病前期患者、2型糖尿病患者、糖尿病性视网膜病变患者(DR)各50例作为病例以及健康人群50例作为对照。运用基因芯片检测技术在3组不同疾病阶段的病例组和健康对照组中各选择5例检测筛选血清差异表达的miRNA,运用实时荧光定量聚合酶链式反应(RT-PCR)方法对4组各45例样本进行验证。结果基因芯片检测结果显示,miRNA-92a家族基因中的miRNA-92a-3p在DR组的表达水平(1 453±432)明显高于健康人群组(1 096±223),P<0.05;RT-PCR验证结果显示,DR组与对照组miRNA-92a-3p的差异具有统计学意义,F值为9.98, P<0.001。RT-PCR验证结果与基因芯片检测结果一致,两者的差异均具有统计学意义。结论 miRNA-92a-3p可能在糖尿病患者发生视网膜病变中发挥一定的作用,需要进一步探索其在糖尿病视网膜病变发病机制中的作用。Objective To explore the expression changes of miRNA-92 a-1-5 p and miRNA-92 a-3 p from miRNA-92 a family in healthy people, pre-diabetes, type 2 diabetes and diabetic retinopathy. Methods We respectively collected 50 patients with pre-diabetes, type 2 diabetes mellitus, and diabetic retinopathy as cases, and 50 healthy people as control. We used MicroRNA array to detect and screen significantly different miRNAs in the serum from 15 patients with different stages of disease and 5 healthy controls. We used RT-PCR to verify the significant microRNAs in 45 samples respectively from the four groups. Results MicroRNA array results showed that the serum expression level of miRNA-92 a-3 p in DR group(1453±432) is significantly higher than that in healthy control group(1096±223), p<0.05. RT-PCR verification results showed that miRNA-92 a-3 p in the DR group and healthy control group was statistically significant with F value of 9.98, P<0.0001. RTPCR verification results further validated the MicroRNA array results. Conclusion Mi RNA-92 a-3 p may play a role in diabetic retinopathy, and further study is need to explore its role in the pathogenesis of diabetic retinopathy.
