摘  要:建立了瓜实蝇快速可视化分子检测方法,为瓜实蝇的快速分子检测提供了有力的技术支持。针对瓜实蝇COⅠ基因设计了1组LAMP引物,通过LAMP扩增,能够将瓜实蝇与其近似种区分,具有良好的特异性;反应灵敏度好,能够检测到1.65 ng/μL的DNA模板;能够通过快速粗提的DNA作为模板进行扩增。从DNA提取开始80 min内完成检测,大大提高了检测速度。同时可视化检测需要的设备简单,操作简便,适用于口岸一线和野外等工作条件下使用。A rapid visual molecular detection of Bactrocera cucurbitae was established.It provided a technical support for rapid molecular detection of B.cucurbitae.A group of primers was designed to distinguish B.cucurbitae from similar species by LAMP.The reaction system had high specificity and sensitivity,by which 1.65 ng/uL was the lowest DNA concentration that could be detected.Moreover,the rapid extraction of DNA could be detected.It would reduce the detection time to less than 80 min from DNA extracting to detection completed.The visual detection just demanded for simple water bath equipment, and was easy to operate,so it would be convenient to apply to the basic laboratories and laboratories without molecular detection equipments.
