摘  要:为了研究四环素(TC)多克隆抗体的制备方法,试验采用碳二亚胺法制备TC完全抗原,将制备好的免疫抗原对昆明鼠进行免疫,免疫结束后对小鼠进行眼眶取血,获得TC多克隆抗体,通过间接酶联免疫吸附试验(ELISA)法测定抗体效价,通过间接竞争ELISA法测定多克隆抗体特异性。结果表明:该多克隆抗体效价达到1∶32 000以上,抗体浓度为4.36 mg/mL;与氯霉素、特布他林、沙拉沙星等常用兽药无交叉反应。说明制备的TC多克隆抗体特异性强。In order to study the tetracycline ( TC) polyclonal antibody preparation,the complete antigen of TC was prepared by carbon imine tetracycline method,and kunming mice were immunized by using immune antigen to obtain the polyclonal antibody of TC. The titer was determined by indirect ELISA, and the polyclonal antibody specificity was determined by indirect competition ELISA. The sesults showed that the titer was above 1 ∶32 000. The antibody concentration was 4. 36 mg /mL. The specificity of polyclonal antibody had no cross reaction with other common veterinary drugs, such as chloramphenicol,terbutaline,and sarafloxacin. The results indicate that the tetracycline polyclonal antibody has a strong specificity.
