摘 要:目的 观察七氟烷对新生大鼠海马组织端粒酶反转录酶(TERT)及神经细胞凋亡的影响,探讨七氟烷导致神经细胞凋亡与TERT表达的关系。方法 将新生SD大鼠随机分为对照组(C组:吸2%纯氧)与七氟烷吸入组(S组:吸入2.3%七氟烷)。体外分离大鼠海马组织,原位缺口末端标记法(TUNEL)染色比较神经元凋亡情况,免疫组织化学染色联合反转录-聚合酶链反应(RT-PCR)比较TERT的表达水平。结果 TUNEL染色显示七氟烷处理后,新生大鼠海马神经元的凋亡显著增加(11.408±2.357比1.324±0.032,P<0.05)。与C组比较,S组中的TERT强阳性、阳性和弱阳性的表达均较低,且差异有统计学意义。PCR结果显示七氟烷处理后,大鼠海马神经元中TERT mRNA的表达水平降低至1/14.281。结论 七氟烷诱导海马组织神经细胞凋亡可能与其下调TERT表达密切相关。Objective To investigate the influence of sevoflurane on the telomerase reverse transcriptase (TERT) expression and hippocampal neuron apoptosis of neonatal rats.Methods Hippocampus nerve apoptosis was observed by terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL), and the TERT expression were detected by immunohistochemistry (IHC) and reverse transcriptase-polymerase chain reaction (RT-PCR).Results TUNEL staining showed a significant increase in apoptosis after sevoflurane treatment (11.408±2.357 vs. 1.324±0.032, P<0.05), the result of IHC and qPCR showed that the expression of TERT in sevoflurane group was decreased significantly.Conclusion The apoptosis of neuron by sevoflurane may be associated with the inhibition of TERT.
