摘　　要：目的:采用HPLC法同时测定武当山地区金银花中绿原酸和木犀草苷含量,为金银花生产适宜区的选择提供理论参考。方法:色谱柱为C_(18)柱(250 mm×4.6 mm,5μm),乙腈为流动相A,0.5%冰醋酸水溶液为流动相B,采用梯度洗脱,流速为1 mL/min,检测波长为350 nm。结果:绿原酸、木犀草苷在5～100μg/mL范围内均呈良好的线性关系;精密度、稳定性、回收率试验结果均良好。武当山地区金银花中绿原酸、木犀草苷含量符合《中华人民共和国药典》标准。结论:所建方法适合金银花中绿原酸和木犀草苷含量测定,结果准确可靠。实验结果初步显示,武当山地区适宜金银花栽培。Objective:To establish the HPLC method for determining contents of chlorogenic acid and galuteolin in Flos Lonicerae,and to provide theory references of selecting appropriate areas for cultivation of L. japonica in Wudang mountain region.Method:A separation was performed on a Symmetry ODS-C18 column(250 mm×4.6 mm,5 μm),acetonitrile as mobile phase A,0.5% acetic acid aqueous solution as mobile phase B with gradient elution,flow rate was 1 mL·min-1,the detection wavelength was 350 nm. Results:The main peak and the nearby peaks were separated well,chlorogenic acid and galuteolin both showed good linear relationship in the range of 5~100 μg/mL. The results of precision,stability and recovery test were good. Contents of chlorogenic acid and galuteolin in Flos Lonicerae in Wudang mountain region were up to the standard of the Chinese pharmacopoeia. Conclusion:This method was sensitive,convenient,accurate and reliable with good precision. The experimental results preliminarily show that the environmen in Wudang mountain region was appropriate for cultivation of L. japonica.
