摘 要:目的 研究周围神经损伤与再生过程中生长相关蛋白 (Growth associatedprotein ,GAP 43 )mRNA表达和蛋白质合成的变化规律。方法 建立大鼠坐骨神经中段钳夹损伤模型 ,用原位杂交和免疫组化技术观察大鼠腰髓和背根神经节的GAP 43mRNA表达和蛋白质合成变化。结果 大鼠坐骨神经损伤后 ,腰髓腹角运动神经元和背根节感觉神经元GAP 43mRNA表达和蛋白质合成增强。结论 周围神经损伤与再生中GAP 43mRNA表达显著增强 ,神经再生完成后表达减弱 ,表明GAPObjective To investigate the expression of GAP 43mRNA and protein in corresponding spinal cord and dorsal root ganglion (DRG) during peripheral nerve regeneration. Methods Using in situ hybridization and immunohistochemistry technique, the expression of GAP 43mRNA and protein in corresponding spinal cord and DRG were investigated after the right sciatic nerve was crushed in adult rats. Results The GAP 43 mRNA expression and protein synthesis were increased in motor neurons of lumbar spinal cord and primary sensory neurons of DRG after nerve injury. Conclusion Expression of GAP 43 mRNA was induced by axotomy and associated with the regeneration state of the neurons. High level of GAP 43 could regulate intrinsic events in neurons which might have significant effect on promoting axon regeneration.
