摘 要:目的 :研究有氧训练对慢性冠状动脉狭窄动物血管内皮生长因子 (vascularendothelialgrowthfactor,VEGF)及心肌侧枝血管生成的影响。方法 :应用Ameroid缩窄器制备小型猪慢性冠状动脉狭窄模型 ,术后 4w随机分为 4组 :对照组 (不运动 ) ,运动 4w组 ,运动 6w组 ,运动 8w组。运动组进行平板训练 :速度 2 .90km/h ,坡度 5 % ,时间30min/d ,5d/w。病理学分析心肌侧枝血管生成 ,Westernblot分析VEGF蛋白表达。结果 :运动组缺血心肌部位的毛细血管数目显著高于非运动组 (6 .9± 1 .3vs 4 .6± 0 .9条 /HPR ,P <0 .0 1 )。VEGF蛋白条带的密度分析显示 :运动 4w组 (652 7± 1 0 89)、运动 6w组 (6739± 1 72 0 )和运动 8w组 (9556± 2 32 9)均显著高于对照组 (2 936± 1 61 0 ,P <0 .0 1 )。运动组合计平均VEGF表达量显著高于非运动组 (782 8± 81 8vs 2 936± 1 61 0 ,P <0 .0 1 )。结论 :有氧训练可以促进慢性冠状动脉狭窄后心肌VEGF表达 。Objective: To study effect and mechanism of vascular endothelial growth factor (VEGF) on collateral development by aerobic training in minipigs with chronic coronary stenosis model. Method: Chronic coronary stenosis model were established by implantation of ameroid occluders on left circumflex coronary artery (LCX) in 18 pigs. Four weeks after surgery, The pigs were divided into sedentary group (n=4) and exercise group (n=8), which further grouped to 4-week (Ex4, n=3), 6-week (Ex6, n=3) and 8-week (Ex8, n=2), respectively. Exercise protocol was 3.8 miles/hour at 5° incline for 30 min/d and 5 d/week. Collateral development was confirmed by pathological analysis. VEGF was analyzed by Western blot. Result: Capillaries in ischemic myocardium in the exercise group were significantly higher than the sedentary group (6.9±1.3/HPR vs 4.6±0.9 /HPR, P <0.01). VEGF in the group Ex4 (6527±1089), Ex6 (6739±1720) and Ex8 (9556±2329) were significantly higher than the sedentary (2936±1610, P <0.01). VEGF expression summed from all exercise groups was higher than the sedentary ( P <0.01). Conclusion: Aerobic training may induce significant myocardial VEGF expression and angiogenesis in pigs with chronic coronary stenosis.
