18β-甘草次酸和甘草酸对人肝癌细胞增殖的抑制和诱导分化作用

摘　　要：目的 :探讨 18β -甘草次酸 (18β -GA)和甘草酸 (GL)对BEL - 74 0 2人肝癌细胞增殖的抑制和诱导分化作用。方法 :用此两种药物处理BEL - 74 0 2细胞 ,测定其对增殖的抑制作用 ,用病理图文分析系统测定细胞的核质比例 ,用RIA法测定AFP分泌量 ,用酶学方法测定细胞内各种酶的比活性。结果 :6 0 μmlo/L、90 μmlo/L、12 0 μmlo/L、18β -GA和 2 5mmlo/L、5mmlo/L、10mmlo/LGL处理细胞 4d后 ,细胞增殖抑制率分别为 32 2 %、5 9 4 %、85 1%和 2 7 7%、6 1 7%、84 8%。 6 0 μmlo/L 18β -GA和 2 5mmlo/LGL处理 4d ,细胞核质比例、AFP分泌量和γ -谷氨酰转肽酶比活性均显著下降 (P <0 0 1和P <0 0 5 ) ,代表细胞分化的鸟氨酸氨基甲酰转移酶、酪氨酸 -a-酮戊二酸转氨酶和碱性磷酸酶比活性则显著升高 (P <0 0 1和P <0 0 5 )。结论 :18β -GA和GL有抑制人肝癌细胞增殖和诱导其分化作用 ,且在同等效果下 18β -GA所需的浓度比GL的浓度低约 4Objective:To investigate the effects of 18 β-glycyrrhetinic acid (18β-GA) and glycyrrhizin (GL) on proliferation inhibition and differentiation induction of BEL-7402 human hepatocracinoma cells. Methods:The cells were treated with 18β-GA and GL,the proliferation and the nucleocytoplasm ratio of the cells were examined,the AFP secretary amount of the cells were determined with radioimmunoassay(RIA),and the specific activities of the intracellular enzymes were assayed by enzymological methods. Results:The cells treated with 18β-GA at dosage of 60μmol/L,90μmol/L and 120μmol/L and GL at 2.5mmol/L,5mmol/L and 10mmol/L for 4 days,the inhibition rates of the cell proliferation were 32.2%,59.4%,85.1%,and 27.7%,61.7%,84.8% respectively.Treated with 60μmol/L of 18β-GA and 2.5mmol/L of GL for 4 days,the nucleocytoplasm ratio of the cells,the AFP secretary amount and the specific activity of γ-GT were all reduced significantly (P<0 01 and P<0 05).While the specific activities of ornithine carbamoyl transferase,tyrosine-α-ketoglutarate transaminase and alkaline phosphatase that they were the differentiated enzymes of the cell were all increased significantly(P<0 01,P<0 05). Conclusion:The 18β-GA and GL can inhibit the proliferation and induce the differentiation of BEL-7402 human hepatocarcinoma cells.In the same effects,the dosage of 18β-GA is lower about 40 times than that of GL.
