摘  要:目的 :探讨IL 15对脐血单个核细胞抗肿瘤活性的影响。方法 :采用MTT法分别检测静止的脐血单个核细胞对K5 6 2的杀伤活性、经不同浓度的IL 15及不同效靶比条件下激活的脐血单个核细胞的抗瘤活性并与成人外周血单个核细胞比较。结果 :静止脐血单个核细胞对K5 6 2的杀伤活性显著低于成人外周血单个核细胞 ,经IL 15活化后脐血单个核细胞抗瘤活性显著增强 ,可达成人水平。本实验中IL 15活化脐血单个核细胞抗肿瘤活性的最佳效靶比为条件为 2 4h ,效靶比为 4 0∶1,最佳浓度为 5 0ng ml。结论 :IL 15在ng水平便能显著提高脐血单个核细胞的抗肿瘤效应 ,为今后优选ILObjective:To identify IL 15 as a novel cytokine that shares many biologic properties with IL 2, and study the efficacy of IL 15 on cord blood mononuclear cell (CBMNC).Methods:CBMNC was incubated with IL 15 at different density or different ratio of effect/target. By using MTT assay to test the specific killing of cord blood.Results:Anactivited citoxicity of fresh CBMNC was significantly lower than that of peripheral blood mononuclear cells.When CBMNC was incubated with IL 15,the level of CBMNC citoxicity was significantly increased and even could reach the level of PBMNC.In our experiment the optimum condition was:24 hours; rIL 15: 50ng/ml;ratio of effect/target:40∶1.Conclusion: IL 15 can significantly increase the citoxicity of CBMNC.Our study suggests that IL 15 can be an alternative cytoking for the adoptive immunotherapy of malignant and nonmalignant disorders.
