碱性成纤维细胞生长因子对庆大霉素诱导的急性肾功能衰竭疗效的探讨

摘　　要：目的 :探讨碱性成纤维细胞生长因子 (b FGF)对庆大霉素 (GM)诱导的急性肾功能衰竭 (ARF)的疗效。方法 :雄性 Wistar大鼠 32只 ,随机分为对照组、实验组 (GM组 )中的 GM1组、GM2组、GM3组各 8只。GM组按 16 0 mg/kg.d- 1肌注 GM8d,对照组注射等体积生理盐水。于实验第 9天从对照组和 GM1组动物腹主动脉取血样做血清尿素氮 (BU N)和血清肌酐 (Scr)测定后 ,经心脏灌注固定 ,取肾皮质做光镜检查。剩下的 GM2组、GM3组动物于实验第 9天停止肌注 GM,GM2组按 b FGF2 0 0 0 U/kg.d- 1 肌注 8d,GM3组则注射等体积的生理盐水 ,之后又于第 9天从 GM2组、GM3组动物中取血样做生化检查、取肾皮质做光镜和电镜观察。结果 :b FGF可促进 GM诱导的损伤肾小管上皮细胞的恢复和再生 ,但血清学变化不明显。结论 :1b FGF对 GM诱导的 ARF有一定的治疗效果 ;2 Scr和 BUObjectives: To explore therapeutic effects of basic Fibroblast Growth Factor (bFGF) in Acute Renal Failure(ARF) induced by Gantamicin(GM) Methods: 32 male Wistar rats were divided randomly into control group and GM1, GM2 , GM3 group of GM groups, 8 animals in each group GM was injected to the animals of GM groups by muscular route for 8 days at a dosage of 160 mg/kg d\+\{-1\} while Saline solution to control group On the 9th day, the animals of GM1 and control group were killed with rat's blood taken for BUN and Scr testing The cortex of kidneys were cut for LM examinations At the same time, bFGF(2 000 U/kg d\+\{-1\}) was injected muscularly to the animals of GM2 group for 8 days while saline solution to GM3 group On the second 9th day of our experiment , the animals of GM2 and GM3 group were killed with the rat's blood taken for BUN, Scr testing and the cortex of kidneys for LM and EM examinations Results: bFGF could promote the recovery and regeneration of renal tubule damaged by GM, but the change of serology was not evident Conclusions: ① bFGF had certain therapeutic effects on ARF caused by GM ② normality of Scr and BUN didn't necessarily mean that there were no pathological changes in kidneys
