体外蒿甲醚伍用血红素对血吸虫磷酸甘油酸激酶和丙酮酸激酶的影响

摘　　要：目的 观察体外蒿甲醚与血红素伍用对日本血吸虫磷酸甘油酸激酶（PGK）和丙酮酸激酶（PK）的影响。方法 以 4～5周龄的血吸虫,置于含蒿甲醚和/或血红素的培素液内培养24h后,测定虫体的PGK和PK酶活力。结果体外,蒿甲醚（50μmol/L）或血红素（50μmol/L）对血吸虫的PGK和PK酶活力无影响,但两者伍用可引起两种酶的活力明显下降。结论 蒿甲醚对血吸虫PGK和PK的影响具有血红素依赖性。ve To study the effect of combining artemether with heme on activities of phosphoglyc-erate(PGK) and pyruvate kinase(PK) in adult Schistosoma japonicum in vitro. Methods The activities oj PGK and PK were measured in paired worms incubated without or with artemether and/ or heme for 24 h in vitro. Results No effect on the activities of PGK and PK was observed in the worms exposed to artemether(50μmol/L)or heme(50μmol/L) alone for 24h. However, the combined chemotherapy led to a significant decrease in the PGK and PK activities in the parasite. Conclusion Artemether showed a heme - dependent effect on the PGK and PK activities in schistosomes.
