摘 要:目的 :观测七叶皂苷钠对大鼠大脑中动脉闭塞缺血半球梗死面积及凋亡细胞数目的影响。 方法 :选用健康雄性 SD大鼠 ,以头端烫圆的 4 - 0尼龙线由左侧颈总动脉插入 ,阻塞左侧大脑中动脉 ,建立缺血 2 h再灌注 2 2 h动物模型。再灌注即刻腹腔内注射七叶皂苷钠 (5 m g/ kg)为治疗组 ,注射生理盐水 (10μl/ g)为对照组。对视交叉部冠状脑组织块行 TTC染色 ,测定其吻端梗死面积百分比 ;对邻近脑组织块行原位末端转移酶标记 (TU NEL ) ,计数缺血半球凋亡细胞数目。 结果 :治疗组和对照组大鼠脑梗死面积分别为 (8.15± 5 .0 ) %和 (2 2 .14± 7.7) % (P <0 .0 1) ;细胞凋亡数目分别为 70± 2 3和 2 2 5± 78(P<0 .0 1)。结论 :再灌注即刻腹腔内注射七叶皂苷钠对大鼠短暂性、局灶性脑缺血具有保护作用 ;Objective: To investigate the effect of sodium aescinate on infarct volume and neuron apoptosis in transient focal cerebral ischemia of rats. Methods: Healthy male SD rats were subjected to middle cerebral artery occlusion for 2 h and reperfusion for 22 h using an intraluminal vascular occlusion method. According to different treatments, the rats were randomly divided into sodium aescinate treated group, saline control group and sham operated group. To estimate the ischemic damage, persentage of the coronal infarct area at the level of the rostral edge of the optic chiasm was calculated with TTC staining. DNA fragmentation was measured in adjacent paraffin sections with terminal deoxynucleotidyl transferase (TdT) mediated dUTP biotin nick end labeling (TUNEL) method. The number of apoptotic neurons in ischemia hemispheres was counted. Results: The persentages of infarct area were (22.14±7.7)% and (8.15±5.0)% in control and treatment groups, respectively ( P <0.01). The number of apoptotic neurons was also dramatically smaller in treatment group than in control group (70±23 vs 225±78, P <0.01). As contrasted with control rats, both the infarct volume and apoptotic cells were reduced in treated rats. Conclusion: These data suggest that abdominal injection of sodium aescinate immediately after reperfusion may attenuate cerebral ischmic damage. This positive effect of escin on focal ischemia may partly depend on the declined number of apoptotic cells.
