摘 要:探讨扶正化瘀方逆转肝纤维化的配伍机制 ,以Wistar大鼠皮下注射CCl4 和高脂低蛋白饲料复合造模 ,重复正交试验设计 ,6因素 (6味中药 )、2水平 (用与不用 )、部分一级交互作用。模成后按L1 6 (2 1 5)正交表随机分组经口给药 4周。检测肝羟脯氨酸 (Hyp) ,血清丙氨酸氨基转移酶 (ALT)、总胆红素 (TBil)和白蛋白 (Alb) ,筛选出有显著作用的 4味药物 ,再按上法用L8(2 7)正交表重复正交试验 ;根据 2次结果 ,以秋水仙碱对照 ,比较单味桃仁 (F药 )与优化复方的作用。结果 :F药降低肝组织Hyp含量的作用显著 ,重在提高肝组织金属基质蛋白酶 1(MMPI)活性 ;丹参 (D)可显著提高血清白蛋白、降低血清TBil含量 ,虫草菌丝 (A)既可显著提高血清白蛋白含量、也能有效降低血清ALT活性 ,七叶胆(C)重在降低血清ALT活性。ACDF复方提高肝MMP1 活性 ,降低肝Hyp含量、血清ALT活性及TBil含量 ,提高血清Blb含量等均显著优于F单味药。结论 :在治疗CCl4 大鼠肝纤维化 ,化瘀药桃仁促进胶原降解 ,降低肝组织HyP含量 ;补虚益精的虫草菌丝、活血养血的丹参保护肝细胞 ,改善肝功能 ;清热解毒的七叶胆抗肝细胞损伤 ;4药配伍后 。In order to explore the combination mechanism of the formula to support the anti pathogenic ability and dissipate blood stasis in promoting reversion of liver fibrosis,Wster rats were medeled by subcutaneous injection of CCI 4 and high fat and low protein forage,for repeating orthogonal test,6 factors (6 herbal ingredients),2 levels (use and disuse) and partial first class interaction.After modeling,the rats were divided randomly upon L 16 (2 15 ) orthogonal table into the groups and were given drugs orally for four weeks to determine hepatic hydroxy proline (Hyp),alanine transaminase (ALT), total bilirubin (TBil) and albumin (Alb) for screening four herbal ingredients with remarkable functions.Then,the orthogonal test was repeated with L 8(2 7) orthogonal table by the above method.In accordance with the results in the two tests,the functions of single Semen Persicas (F drug) and the perfected compound formula were compared with colchicines.The results showed that the function of F drug in decreasing Hyp content in the hepatic tissues was remarkable,in a stress to elevate the activity of the hepatic matrix proteolytic enzyme (MMP 1).And Radix Salvia Miltiorrhiziae could remarkably elevate Alb and decrease Tbil contents,and cordycepic hyphae (A) could remarkably elevate Alb content and decrease ALT activity effectively,and Gynostemma pentaphylla (C drug) could decrease ALT activity predominantly.The function of ACDF compound formula could elevate the activity of hepatic MMP 1,decrease the hepatic Hyp content,ALT activity and Tbil content and elevate ALB content,better than single F drug.The conclusion is that in the treatment of liver fibrosis in CCI 4 rats,Semen Persicae that can disperse blood stasis promotes collagen degradation and decreases the hepatic collagen content.Cordycepic hyphae that can tonify deficiency and benefit essence and Radix Salvia Miltiorrhiziae that can activate and nourish blood protect the hepatic cells and improve the hepatic functions.Gynostemma pentaphylla that can c
