摘 要:目的 研究脑组织中白细胞介素 - 1β(IL - 1β)、神经酰胺、caspase - 3在创伤性脑损伤 (TBI)后的改变和相互作用。 方法 以大鼠自由落体脑损伤为模型 ,损伤前后各 30min加用不同处理因素后 2 4h ,用干湿法测量脑含水量 ,TUNEL染色检测神经细胞凋亡 ,ABC法检测caspase- 3活性片段P2 0的表达 ,DAG激酶 (diacylglycerolkinase ,DAGK)法检测神经酰胺的变化。 结果 TBI后双侧脑组织含水量、TUNEL染色阳性细胞、caspase- 3活性片段、IL - 1β及神经酰胺均较对照组显著增加。caspase - 3抑制剂使TBI后TUNEL染色阳性细胞明显下降 ,对脑组织水和神经酰胺含量没有影响。IL - 1β抗体使TBI后脑组织中神经酰胺含量显著下降 ,同时也使TBI后脑组织含水量、TUNEL染色阳性细胞、caspase - 3活性片段下降 ,但明显弱于神经酰胺的降解剂鞘氨醇激酶(SK)的作用。而SK对TBI后脑组织中IL - 1β的表达没有影响。 结论 TBI后脑组织中神经酰胺的大量合成在伤后的脑水肿和神经细胞凋亡中是一个主要信号途径。IL - 1β是神经酰胺合成的刺激因素之一 ;Objective To study the changes and effects of ceramide, interleukin 1β(IL 1β) and caspase 3 in cerebrum after traumatic brain injury (TBI). Methods TBI models of rats were made with Feeney's method, and caspase 3 inhibitor z DEVD fmk,IL 1β antibody and ceramide degradation reagent sphingosine kinase(SK) were administered by intraventricular microinjections. The changes of water content, number of apoptotic cells, levels of IL 1β, caspase 3 and ceramide in brain tissues were monitored at 24 hours after TBI. Results After TBI, bilateral cerebral water content, TUNEL positive neuronal cells numbers, caspase 3, IL 1β, and ceramide expression in cerebrum increased significantly, z DEVD fmk decreased the number of TUNEL positive neuronal cells induced by TBI, and there was no effect on the production of ceramide and IL 1β. IL 1β antibody reduced the content of ceramide in cerebrum after TBI. It also decreased water content, the number of apoptotic cells, and caspase 3 in brain tissues after TBI, but its effects is not as notably as the effects of SK, and SK did not effect on IL 1β inducing in cerabrum after TBI. Conclusions The massive synthesis of brain ceramide is a very important medium in the secondary brain injury after TBI, and IL 1β is one of the factors that stimulate ceramide synthesis.
