摘　　要：目的　观察生物半透膜 (BSM)包裹胰岛样细胞团 (ICCs)与富含Sertoli细胞的睾丸样细胞团 (SC)体外培养时 ,Sertoli细胞是否同时表达CD95L和TGF β基因及体外抗免疫排斥作用的效果。方法　BSM包裹猪ICCs和SC ,与人淋巴细胞进行培养。免疫组化染色观察CD95L、TGF β的表达。通过测定胰岛素浓度和刺激释放实验 ,AO PI荧光双染色和TUNEL染色 ,观察培养细胞的结构功能状况。结果　ICCs和SC共同培养时 ,许多Sertoli细胞表达CD95L和TGF β蛋白。与淋巴细胞培养后 ,胰岛素分泌良好 ,刺激释放实验比值 >2 0 ,AO PI荧光染色超过 95 %的细胞存活 ,TUNEL染色未发现凋亡细胞。结论　Sertoli细胞与胰岛细胞共同培养时可出现CD95L和TGFObjective To identify the coexpressions of CD95L and TGF β in Sertoli cells (SC) and the effect of decreasing the rejection in vitro when the islets and SC were enwrapped with biological semipermeable membrane (BSM). Methods The ICCs(islet like cluster cells) and SC of newborn porcine were enwrapped with BSM, and cocultured with lymphocytes. The contents of insulin were measured. The DNA fragmentations, the viability of ICCs and the expressions of CD95L and TGF β genes were observed. Results There were coexpressions of CD95L and TGF β in Sertoli cells when cocultured with islets. The TUNEL staining did not reveal any apoptotic cells. The insulin secretion was excellent in the two groups. Conclusion The coculture of ICCS with SC could provide a micromeliu of coexpression of CD95L and TGF β for decreasing the rejection in vitro .
