摘 要:目的 :探讨导电聚吡咯膜细胞培养技术是否会改变细胞的生物学特性。方法 :应用形态学观察、分子荧光探针及划痕标记染料示踪技术 ,在细胞形态学、超微结构、细胞膜流动性以及细胞间通讯连接功能等方面 ,对采用导电聚吡咯膜细胞培养技术和常规培养技术培养的大鼠角朊细胞的生物学特性分别进行检测与比较。结果 :使用导电聚吡咯膜细胞培养技术培养的大鼠角朊细胞 ,在细胞形态学、超微结构、细胞膜流动性以及细胞间通讯连接功能等方面 ,与常规方法培养的大鼠角朊细胞相比未观察到异常改变。结论 :在现有实验条件下 。Objective To study the biology characterizations of rat keratinocytes based on electrically conducting polypyrrole cell culture technique.Methods The rat keratinocytes were isolated and cultured with electrically conducting polypyrrole cell culture technique and classical technique,respectively.The cell morphology was studied with optical,fluoresc^ence and electron microscopes.Cell membrane fluidity was measured by fluorometry and cell gap junctional intercellular commu^nications by scrape loading assay.Results The microscopic study showed no obvious changes in size,shape,nucleus size and cell ultra structure in rat keratinocytes based on electrically conducting polypyrrole cell culture technique compared with those based on classical culture technique.No significant differences in cell membrane fluidity and cell gap junctional intercellular communications were observed.Conclusion The cell biology characterizations of rat keratinocytes based on conductive polypyrrole cell culture technique has no significant changes copaired with the normal keratinocytes.
