摘  要:探讨热休克蛋白 70 (Hsp70 ) - H2 2肽复合物与小鼠骨髓来源的树突状细胞 (dendritic cell,DC)成熟之间的关系。采用 GM- CSF、IL- 4刺激培养小鼠骨髓细胞 ,增殖产生大量 DCs;从 H2 2肝癌细胞中获取肿瘤细胞肽 ,体外与Hsp70进行结合 ,结合产物修饰 DC,通过细胞因子检测试剂盒和流式细胞仪对修饰的 DCs上清液和 DCs膜表面分子进行检测。发现 Hsp70 - H2 2肽复合物修饰的 DCs大量分泌 IL- 12、TNF- α、IL- 1β等细胞因子 ,并且高表达主要组织相容性 类抗原 (MHC 类 )、 CD80、 CD40等分子 ,而单独的 Hsp70或 H2 2肽则不具有此作用。结果提示 :Hsp70 - H2 2肽复合物可诱导 DC成熟 ,这种成熟的The relationship between Hsp70-H22 peptide complexes and maturation of marrow-derived dentritic cells (DC) was studied. Murine bone cells were cultured with GM-CSF and IL-4 stimulating, thus proliferated to produce plenty of DCs. The tumor antigen peptides obtained from H22 liver cancer cells bound with Hsp70 in vitro. After DCs were modified by Hsp70-I22 peptide complexes DC supernatant and DC membrane surface molecules were detected using cytokine-detected kit and flow cytometry. The findings indicated that DCs modified by Hsp70-H22 peptide complexes could plentifully secret IL-12, TNF-α, IL-1β and highly express MHCⅡ antigen, CD80, CD40 on surface, but single Hsp70 or H22 peptide didn't have the above effect. It was suggested that Hsp70-H22 peptide complexes could induce the maturation of DCs and the secreted cytokines and expressed molecules by DCs were important in antitumor course.
