摘  要:目的 观察转化生长因子 β1 (TGF- β1 )基因能否转入骨髓基质干细胞 (MSCs) ,并探讨经 TGF-β1 基因转染后的MSCs增殖和分化情况 .方法 取新西兰雄性大耳白兔的骨髓 ,得骨髓源 MSCs体外培养 .将重组 TGF-β1 和空载 pc D-NA3基因分别转染 MSCs后 ,免疫组化 (SABC)法检测 TGF-β1 转染是否成功 ,用透射电镜和流式细胞仪观察两组 MSCs的增殖和分化情况 .结果  SABC法证明 TGF-β1 瞬间转染成功 ;转染 4 8h后 ,实验组 MSCs增殖较对照组显著 ;实验组MSCs具有一定的分化趋势 .结论 重组 TGF-β1 基因转入骨髓源 MSCs的方法是可行的 ,瞬间 TGF- β1 转染对AIM To investigate whether transforming growth factor β 1 (TGF β 1) gene can be successfully transfered into mesenchymal stem cells (MSCs)and if it can be, then to observe the effect of the successful recombinant TGF β 1 gene transfer on MSCs. METHODS Adult male New Zealand rabbits were used to collect MSCs. After the transfection of both recombinant TGF β 1 and pcDNA3 genes, the multiplication and differentiation of MSCs were observed by flowcytometry and transilluminating electron microscope and the positive expression of TGF β 1 was detected by immunohistochemical staining (SABC). RESULTS Transfer was successful with the evidence of SABC. There was significant multiplification in MSCs with TGF β 1 transfer and the trend of the differentiation was obvious after transient transfection. CONCLUSION TGF β 1 gene can be successfully transferred into MSCs. 48 hours after the transfection, the MSCs number increased obviously and the differentiation of MSCs is significant.
