摘 要:采用糖尿病大鼠模型 ,以核因子 (NF) - κB亚基 P6 5多抗为抗体 ,应用免疫组织化学染色法观察氯沙坦(losartan)对糖尿病大鼠肾脏 NF- κB表达的影响 ,并进一步分析 NF- κB表达与肾小球内粘附分子 (ICAM- 1)蛋白表达、单核细胞浸润及肾脏功能和结构损害的关系。结果表明 :糖尿病大鼠肾小球 NF- κB表达较正常肾组织增高 ,氯沙坦可显著降低 NF- κB的表达。且 NF- κB阳性细胞数与肾小球 ICAM- 1蛋白表达、单核细胞浸润、肾脏功能和结构损害显著相关。结果提示氯沙坦对糖尿病大鼠肾脏保护作用可能通过抑制血管紧张素 (Ang )引起的 NF- κB活化 ,下调受NF- κB调控的 ICAM- 1表达 。By using the model of diabetic rats, the effects of Losartan on renal NF-κB P65 expression in diabetic rats were studied by using immunocytochemical techniques using NF-κB P65 as antibody. At the same time, the relationship between glomerular expression of NF-κB and ICAM-1 expression, macrophage accumulation in diabetic rats was examined. The results showed that expression of NF-κB in glomerulus of diabetic rats was higher than in the normal control, while Losartan could decrease the expression of NF-κB in diabetic rats. The number of positive cells expressing NF-κB in glomerulus of diabetic rats showed a strong correlation with ICAM-1 expression, macrophage accumulation and urine protein excretion, respectively. It suggested that Losartan mihgt protect diabetic renal tissue by inhibiting angiotensin-stimulated NF-κB activation, therefore downregulating ICAM-1 expression and macrophage infiltration which might play a key role in diabetic nephropathy.
