摘　　要：本研究建立了简单快速抽提新生隐球菌基因组DNA的方法,适用于PCR和酶切分析。方法是细胞悬浮于含5mmol/LDTT的1%Sarkosyl抽提缓冲液中,通过加入玻璃珠在混漩器上剧烈振荡破碎细胞壁和荚膜;然后酚氯仿抽提DNA,1×108细胞约可得1μgDNA。In this study, a simple and rapid method for extracting genomic DNA from Cryptococcus neoformans was established and used as template for PCR and restriction endonucleases analysis. The cells(containing cell wall and polysaccaride capsule) were suspended in 1% sarkosyl extraction buffer containing 5mmol/L DTT and broken by vigorously mixing with glass beads on a votorex.DNA was extracted with phenol chloroform .About 1μg DNA was obtained from 1×108 cells.
