摘 要:目的;探讨一种新的化合物肉桂酸锗(肉桂酸锗氮桥环化合物)不同给药途径对小鼠宫颈癌14号(U14)的抑制作用。方法:通过体内实验,观察肉桂酸锗水溶剂灌胃及腹腔注射两种给药途径对荷瘤U14小鼠实体瘤生长的影响(抑制率)及病理形态改变,凋亡细胞百分率。结果:肉桂酸锗水溶剂灌胃组与腹腔注射组小鼠瘤重明显减轻、抑制率分别为50.25%与43.72%,明显高于模型空白组(P<0.05)。甲基绿-派诺宁染色显示瘤细胞有凋亡现象,灌胃组瘤细胞凋亡百分率为(8.3±2.7)%;腹腔注射组瘤细胞凋亡百分率为(6.5±2.3)%,与模型空白组(3.3±1.6)%相比,有显著性差异(P<0.01),且肉桂酸锗组各主要脏器未见明显毒性反应。结论:肉桂酸锗对小鼠U14瘤的生长有明显抑制作用,其机理可能是诱导瘤细胞凋亡。ve:Discuss the inhibitory effect of a novel compound germanium cinnamate(germanium cinnamate nitrogen bridge eyelic compound) against Cervix Cancer No. 14 in mice at different administration pathway. Methods In vivo, observe the effect of germanium cinnamate water solution on growth(inhibitory rate), pathological morphology and apoptosis cell percentage of the entitative tumor in U14 tumor loading mice via stomach lavage and abdominal injection. Results: The tumor weights of groups germanium cinnamate water solution administrated via stomach lavage and abdominal injection were reduced obviously, the inhibitory rate was 50. 25% and 43. 72% respectively, higher than the control(P<0. 05). Methyl green-pyronin staining indicated that the tumor cells had apoptosis phenomenon, the percentage of apoptosis of tumor cells was (8. 3 ± 2. 7)% in stomach lavage group, (6. 5 ± 2. 3)% in abdominal injection group, compare to(3. 3 ±1. 6)% in control, had remarkable difference(P< 0. 01). Also, obvious toxic reaction were not seen in all main organs of germanium cinnamate groups. Conclusion: Germanium cinnamate had obvious inhibitory effects on growth of UM in mice, the mechanism may involve in inducing apoptosis of the tumor cells.
