利福霉素B产生菌的推理选育

摘　　要：利福霉素 B由地中海拟分枝酸菌产生，本文对工业生产菌株A．mediterranei XC1-02进行进一步筛选。采用推理选育的方法使产生菌减轻由芳香族氨基酸[色氨酸（trp）、苯丙氨酸、酪氨酸]和对羟基苯甲酸（PHBA，Phb）引起的对利福霉素B生物合成的反馈抑制作用，并提高对前体丙酸（prp）的耐受量。通过UV诱变提高诱变株的耐受性，获得了高产菌株A． mediterranei XC 9-25（trpt，phbt prpt），其生产能力达到 1000u/ml，较原始出发菌株A．mediterranei XC 1－02提高了 1．385倍。动力学试验结果表明利福霉素B的生物合成与菌体生长不同步，属于非生长偶联型。ABSTRACT:Rifamycin B is produced by Amycolatopsis mediterranei. A working strain A. mediterranei XC 1-02was used for further screening, and specific mutation and screening procedures were adopted to select a strain, whichcan alleviate the inhibition caused by both aromatic amino acid and PHBA in the metabolic pathway of rifamycin B aswell as enhance the production of propionate which is one of the precursors of rifamycin B biosynthesis. Finally a strainA. mediterranei XC 9-25 was obtained, and its rifamycin B productivity reaches 10000u/ml, which was 1. 38 times high-er than that of the parent strain XC 1-02. The analysis of kinetic experiments indicated that the biosynthesis of rifamycinB and the growing rate of the strain was nonsynchronized.
