摘 要:本文通过正交分析,以最终收率和效价为指标对妥布霉素结晶过程中的影响因素如:乙醇添加量、温度、时间、搅拌速度等进行了研究并对其优化,得出了不同情况下的最佳参数组合:当考虑收率时,采用A3B2C3D2组合即:结晶液:乙醇为1: 12;结晶温度为25°C,结晶时间为14h,搅拌速度为200r/min。当考虑效价时,最佳影响因素组合A2B3C1D2,即:结晶液:乙醇为1:10;结晶温度为35℃,结晶时间为10h,搅拌速度为200r/min。综合平衡后可用作为实际生产的最佳影响因素组合,结晶液:乙醇1:12,结晶温度25℃,结晶时间10h,搅拌速度300r/min。ABSTRACT:In this paper, the effective factors such as ethanol addition volume, temperature, time and stirringrate and so on in the process of crystallization of tobramycin with the final yield and titer as indices was analysed byorthogonal method. At the same time, the optimal reference combination under different conditions was obtained. Whenthe final yield was put in the first place, the optimal condition was as follows: crystallization solution: ethanol = 1: 12,temperature 25°C, time 14h and stirring rate 200r/min; When the titer was considered in the first place, the optimalreference combination was as follows: crystallization solution: ethanol = 1: 10, temperature 35°C, time 10h and stirring rate200r/min. Considering comprehensively, the optimal condition was as follows: crystallization solution: ethanol = 1: 12, temperatuer 25°C,time 10h and stirring rate 300r/min.
