UVC与油酸钠联用消毒对血浆有效成分的影响

摘　　要：为了解可有效灭活血浆中脂包膜病毒的ＵＶＣ与油酸钠联合作用剂量（３２００μＷ／ｃｍ２ＵＶＣ＋０．３％油酸钠作用３０ｍｉｎ或３２００μＷ／ｃｍ２ＵＶＣ＋０．２％油酸钠作用４０ｍｉｎ）对血浆有效成分的影响，对经ＵＶＣ与油酸钠处理后血浆与对照血浆分别进行了生化、微量免疫电泳、交叉免疫电泳、ＳＤＳ—ＰＡＧＥ、Ｗｅｓｔｅｒｎ—Ｂｌｏｔ等分析。结果，处理后血浆中ＩｇＧ以及主要蛋白质的分子量都无明显变化，但少量蛋白分子有多聚体形成；４种酶的活性，除碱性磷酸酶外，乳酸脱氢酶、天冬氨酸转氨酶、丙氨酸转氨酶均明显下降；ＦⅧ促凝活性仅为对照组的２３．０％～２４．５％，活化部分凝血酶时间延长超过３倍，凝血酶原时间延长超过４倍。因此，该方法对临床直接使用的血浆、白蛋白及免疫球蛋白制品的消毒是可行的，但不能用于消毒生产凝血因子和血浆酶制剂的原料血浆及凝血因子类制剂。In order to know the influence of the combined disinfection dose of UVC and sodium oleate effective for inactivating the viruses with lipid envelope in plasma (3200 μW/cm2 UVC+0.3% sodium oleate with a 30 minute contact time or 3200 μW/cm2 UVC+0.2% sodium oleate with a 40 minute contact time) on the active principles of plasma, analyses, such as biochemistry, microimmunoelectrophoresis, crossed immunoelectrophoresis, SDS-PAGE and western-Blot, of the plasma treated with UVC and sodium oleate and the control plasma were carried out. The results showed that the molecular weight of IgG and main proteins in plasma had no obvious change, but a few protein mole- cules formed polymers. Except alkaline phosphatase, the other enzymes in plasma including lactate dehydrogenase, aspartate transaminase and alanine transaminase decreased obviously. The coagulative activity of F Ⅷ was only 23.0%-24.5% of that in the control group, acti- vated partial thromboplastin time was prolonged by more than three times and prothrombin time was prolonged by more than 4 times. Therefore, this method is feasible for disinfection of plasma, albumin and immunoglobulin products used directly in clinical work, but it can not be used for disinfection of raw plasma in production of coagulation factors and plasma enzyme preparations and coagulation factor preparations.
