摘 要:目的 探讨陕西人群 HUMARA基因多态性分布及与衰老的相关性 .方法 应用聚合酶链反应 (PCR- STR)技术先后对老年人 (92例 )和青年人 (6 3例 )的 HU MARA基因的多态位点进行检测 .结果 在老年组中 ,片段大小在 2 80~4 4 5 bp之间 ,重复片段为 (GCA) ,重复次数最少 2 2次最多 77次 .频率分布在 0 .0 0 5~ 0 .39,高峰在 30 0~ 310 bp(39% ) ,杂合率为 0 .83,多态信息量 (PIC)为 0 .76 ;在青年组中 ,片段大小在 2 80~ 4 0 8bp之间 ,重复次数最少 2 2次最多 6 5次 ,频率分布在 0 .0 1~ 0 .33,高峰在 310~ 32 0 bp(33% ) ,杂合率为 0 .90 ,多态信息量 (PIC)为 0 .83.老年组基因频率分布与青年组比较 ,有显著性差异 (P<0 .0 1) .结论 推测该位点遗传多态与细胞的衰老有一定的相关性 。AIM To investigate the polymorphism of the HUMARA gene in Shaanxi population of China and its relationship to aging. METHODS Polymorphism sites of the HUMARA gene in 92 cases of unrelated aged people and 63 cases of unrelated youth in Shaanxi were detected by PCR STRmethod. RESULTS The allele fragments ranged from 280 to 445 bp in the aged. The repetitive fragment was (GCA) and repetitive fraguency was 22 to 77. The distribution of allele frequency was 0.005 to 0.39. The heterozygosity rate was 0.83 and polymorphism information content (PIC) was 0.76. The allele fragments ranged from 280 to 408 bp in the youth. The repeat number was 22 to 65. The distribution of allele frequency was 0.01 to 0.33. The heterozygosity rate was 0.90 and PIC was 0.83. The distribution of polymorphism of the HUMARA gene varied significantly between the aged and the youth ( P <0.01). CONCLUSION The relationship between the HUMARA gene and aging needs further study.
