摘  要:目的 观察氟伐他汀 (fluvastatin)对博来霉素(bleom ycin A5)所致的大鼠肺纤维化的治疗作用 ,为肺纤维化的治疗探索一种新的方法 .方法 雄性 SD大鼠 30只 ,随机分为正常对照组 (C组 )、博来霉素组 (B组 )及根据给博来霉素后第 1,15日始每日按 2 0 m g·kg- 1及 10 mg· kg- 1给予氟伐他汀灌胃治疗分为 4个治疗组 (TH1,TL1,TH15及 TL15组 ) .2 8d后处死所有动物 ,提取肺组织 ,HE染色 ,显微镜下观察 ,L eica Q5 0 0 MC图像分析系统测定肺泡间隔成纤维细胞数量、厚度及肺间质面积 .Masson、苦味酸天狼猩红染色 ,观察基质胶原及 , 型胶原变化 .肺组织匀浆测定羟脯氨酸含量 ,并比较各组间变化 .结果  1B组的肺泡间隔成纤维细胞数量、厚度、肺间质面积及肺组织羟脯氨酸含量均高于 C组 (P<0 .0 1) ,各氟伐他汀治疗组的以上各观察指标均低于 B组 (P<0 .0 1或 P<0 .0 5 ) .2肺泡间隔成纤维细胞数量、厚度、肺间质面积在 TH1组低于 TH15组 (P<0 .0 1) ,TL1组低于 TL15组 (P<0 .0 1或 P<0 .0 5 ) .3B组基质胶原及 , 型胶原分布较 C组明显增多 ,各氟伐他汀治疗组均有不同程度的减少 .结论 氟伐他汀对博来霉素所致的大鼠肺纤维化具有明显治疗作用 。AIM To investigate the therapeutic effect of fluvastatin in bleomycin-induced pulmonary fibrosis of rats. METHODS Thirty SD rats were divided into six groups: The normal control group (Group C), the model group treated with bleomycinA 5 (Group B) and the rest (Groups T H1, T H15 , T L1 and T L15) so named according to the dose of fluvastatin which were 20 mg·kg -1 and 10 mg·kg -1 and the date which were the first and the fifteenth day after the rats were given bleomycinA 5 respectively. All the rats were killed on the twenty-eighth day. The number of fibroblast and the thickness of alveolar wall, the area of mesenchyma in lung tissue were measured by HE staining. The extracellular matrix and Ⅰ, Ⅲ type collagen in lung tissue were observed by Masson and sirius red staining and hydroxyproline was measured in lung tissue homogenates. RESULTS The number of fibroblast and the thickness of lung alveolar wall, the area of mesenchyma and the content of hydroxyproline in lung tissue were significantly higher in Group B than in Group C (P<0.01). The data in the treatment groups were lower than those in Group B (P<0.01 or P<0.05). It was also found that the data of HE staining were lower in Group T H1 than in Group T H15 (P<0.01) and in Group T L1 than in Group T L15 (P<0.01 or P<0.05). In addition, the extracellular matrix and Ⅰ, Ⅲ type collagen in lung tissue were more in group B than in group C, and compared with group B, they were less in the treatment groups. CONCLUSION Fluvastatin might be a therapeutic medicine for pulmonary fibrosis.
