摘  要:目的探讨IL-12和IL-12受体(IL-12R)在弓形虫感染致不良妊娠结局中的作用机制。方法将24只C57BL/6孕鼠随机分为感染组(n=12)和对照组(n=12),感染组小鼠于孕8d每只腹腔注射400个RH株刚地弓形虫速殖子,对照组注射等量无菌PBS,所有孕鼠均于孕14d时安乐死,观察胎盘及胎鼠的发育情况,采用流式细胞仪的方法检测小鼠子宫自然杀伤细胞(uNK)表面的IL-12受体表达水平的高低,用ELISA的方法检测小鼠胎盘分泌的IL-12细胞因子水平的高低。结果在孕14d感染组小鼠的胎盘出血坏死较多,未感染的对照组小鼠胎盘血供丰富。感染组小鼠的死胎率比对照组小鼠显著增加(P<0.01)。弓形虫感染组孕鼠子宫自然杀伤细胞表面的IL-12受体表达水平明显高于正常对照组(P<0.05),感染组孕鼠胎盘中的IL-12水平明显高于正常对照组(P<0.05)。结论 IL-12水平升高和IL-12受体的高表达在弓形虫感染致不良妊娠结局中发挥重要作用。Objective To investigate whether IL-12 and IL-12 receptor (IL 12R) are related with adverse pregnancy outcomes induced by Toxoplasma gondii. Methods Twenty-four pregnant C57BL/6 mice were randomly divided into infection group (n = 12) and control group (n= 12). Infection group mice were inoculated intraperitoneally with 400 tachyzoites on GD 8 and con- trol group mice were inoculated with same dose sterile PBS at the same time. All mice were euthanized on GD 14,placentas and fetuses were observed. IL-12 receptor levels expressed on mouse uterine natural killer cells (uNK) were detected using flow cy- tometry,and IL 12 levels secreted in placentas were measured by ELISA. Results On GD 14, we found that the placentas from infected mice were hemorrhagic when compared to those of uninfected control mice,which had a better blood supply. The still- birth rate of infected mice was significantly increased compared with that of control mice (P〈0.01 }. The levels of IL-12R ex- pressed on infected uterine NK cells were significantly increased relative to uninfected controls (P〈0. 05) ,and the levels of IL- l2 were higher in samples from the infected group than from controls (P〈0.05). Conclusion The elevated expression levels of IL-12 and IL-12 receptor play an important role in adverse pregnancy outcomes caused by Toxopiasma gondii.
