摘  要:目的 研究类缺血状态下神经元胞质内游离钙离子及细胞活性变化 ,探讨缺血性神经元的损伤与细胞内钙离子的关系及西比灵的治疗意义 .方法 利用 Ca2 +指示剂 Flu- 3/AM作为细胞内钙离子的荧光探针负载培养的神经元 ,共聚焦技术检测细胞内荧光强度的变化 ,利用四唑蓝 (MTT)比色试验检测缺血性神经元的损伤程度 .结果 给予神经元类缺血处理 10 min后 ,缺血组神经元胞质内钙离子浓度和细胞损伤程度明显高于西比灵预处理组 (P<0 .0 1) .结论 西比灵可明显抑制神经元胞质内钙离子的升高 。AIM To study the changes of intracellular Ca 2+ and cell activity and protection of Sibelium on cortical neurons on ischemic-like condition. METHODS Neurons were cultured and loaded with Flu-3/AM, I was measured by fluorescent intensity (FI) in each cell with confocal microscopy, and the extent of neurons injury was analyzed with MTT reduction assay. RESULTS After 10 min ischemic-like conditioning, a rapid increase in fluorescence intensity was observed, there was obvious difference between ischemicgroup and Sibelium treatment group, the fluorescence intensity in ischemic group was higher than that in Sibelium treatment group (P<0.01). The A value of injured neurons in ischemic group was significantly lower than that in therapy group (P<0.01). CONCLUSION Sibelium could reduce the increase of I in cortical neurons and the extent of the injured neurons after ischemic-like conditioning.
