摘  要:获得品质优良的愈伤组织是野生稻资源离体保存和遗传转化的基础。以高秆野生稻为材料,通过采用N6D与NB为基础培养基,研究不同浓度激素组合对高秆野生稻愈伤诱导的影响,并分析比较优质胚性愈伤组织分化成苗的情况。结果表明,以N6D为基础培养基,2,4-D浓度为2.0 mg·L-1的培养基诱导高秆野生稻的出愈率最高,达到50.00%。以NB为基本培养基,最优激素组合为2.0 mg·L-12,4-D+3.0 mg·L-1NAA+3.0 mg·L-1IAA+1.5 mg·L-1KT,出愈率为35.0%,愈伤直径为0.43 cm,愈伤色泽淡黄,质地紧密。说明适宜的激素组合能够诱导高质量的高秆野生稻胚性愈伤,良好的胚性愈伤在MS分化培养基上的分化率为60.0%。通过培养基改良的方式可建立高秆野生稻脱分化与分化的离体研究体系。The cllus culture of wild rice supports conservation in vitro of wild rice and genetic transformation. Cal]us culture of O. alia Swallen was placed on N6D and NB medium containing different hormones. The results showed that the callus ratio of N6D medium with 2mg · L - 12,4-D was 50.0 %, which was the highest callus ratio among all mediums with different hormones. The optimal combination and con- centration of hormones was NB medium with 2.0mg · L^-1 2,4-D + 3.0mg · L^-1 NAA + 3.0mg · L- 1 IAA + 1. S mg · L^-1 KT, under which the callus ratio was 35.0 %, but the diameter of callus was 0.43 cm, and it was tight and yellowish. The callus became green plant by dedi- fferentiation progress, and the green plant regeneration rate was 60 %. Through the improved medium, a differentiation and dedifferentiation in vitro callus culture system of O. a/ta Swallen was established.
