摘 要:以天目铁木嫩茎为外植体,应用均匀设计法筛选其基部直接再生丛生芽苗和试管苗保存的最适合培养基。结果表明,最适合的嫩茎尖基部再生芽苗培养基为:1/4 DR+TDZ 3.80 mg/L+NAA 0.03 mg/L+KT 1.50mg/L,诱导率达99.0%以上;试管苗保存培养基为:1/8 DR+TDZ 0.45 mg/L+KT 1.60 mg/L+根皮苷2.80 mg/L,在试管内保存18个月,平均生长率为2.26%。成功建立了天目铁木嫩茎基部直接再生丛生芽苗和试管苗保存体系。In this study, the tender stem of Ostrya rehderiana was used as explant and the suitable medium compositions were screened for multiple shoots at base of tender stem and in vitro conservation of plantlets by using uniform design experiments. The results showed that the most suitable culture mediums were as following: 1/4 DR + TDZ 3.80 mg/L + NAA 0.03 mg/L + KT i. 50 mg/L for bud seedling regeneration at base of tender stem, the rate of induction was 99.0% ; 1/8 DR + TDZ 0.45 mg/L + KT 1.60 mg/L + Phloridzin 2.80 rag/L, the rate of average growth was 2.26% with 18 month in vitro conservation. Multiple shoots at base of tender stem and in vitro conservation of plantlets of O. rehderiana had been successfully established.
