摘  要:目的:观察通心络对体外模拟脑缺血再灌注损伤大鼠脑微血管内皮细胞的保护作用。方法:分离大鼠脑微血管内皮细胞培养鉴定,采用氧糖剥夺法建立模拟体外脑缺血再灌注内皮细胞损伤模型,利用制备的通心络含药血清干预,CCK-8法检测细胞活性;硝酸酶还原法测定细胞上清液NO的含量;Elisa法检测细胞上清液vWF的含量。结果:与正常组比较,模型组细胞OD值明显降低(P<0.01),细胞上清液NO和vWF(P<0.05或P<0.01)含量明显升高;与模型组比较,10%的通心络含药血清作用24 h,细胞OD值明显增高(P<0.01),细胞上清液NO含量和vWF含量明显降低(P均<0.01)。结论:通心络含药血清可能是通过减少NO和vWF分泌保护脑缺血再灌注损伤的脑微血管内皮细胞。Objective : To observe the protective effects of Tongxinluo on brain microvascular endothelial cells of rats with simulated invitro cerebral ischemia reperfusion injury. Methods: Rat's brain microvascular endothelial cells were isolated, cultured and identified, and the model of simulated in - vitro cerebral ischemia reperfusion injury was established. Then the model was intervened by serum containing Tongxinluo, and its cell viability was determined with CCK - 8 method ; The content of cell supernarant NO was determined by the method of nitrate reductase; The content of cell supernatant vWF was determined by Elisa method. Results: Compared with the normal group, cell's OD value in the model group significantly decreased( P 〈0.01 ) , while NO and vWF content obviously increased(P 〈 0. 05 or P 〈 0.0O1 ). Compared with the model group, 'after 24 hours of using 10% serum containing Tongxinluo, cell's OD value was significantly higher(P 〈 0. 01 ), while NO and vWF content obviously was significantly lower( P 〈 0.01 ). Conclusion: The serum containing Tongxinluo could protect cerebral ischemia- reperfusion injury of brain microvascular endothelial cells by reducing NO and vWF secretion.
