广东汉族人群MMP-9基因R279QR668Q多态性与社区获得性肺炎遗传易感性的关系

摘　　要：目的了解广东汉族人群基质金属蛋白酶-9（MMP～9）基因R279Q、R668Q位点多态性与社区获得性肺炎（CAP）遗传易感性和MMP-9表达水平的关系。方法纳入广东汉族人CAP患者100例为病例组，同期门诊体检的健康广东汉族人96例为对照组，采用酶联免疫吸附测定（ELISA）方法检测病例组发病期、治疗后与对照组的血清MMP-9水平，应用聚合酶链反应-限制性片段长度多态性（PCR—RFLP）方法检测两组MMP-9基因R279Q、R668Q位点的基因型及等位基因。结果病例组发病期的血清MMP-9水平明显高于治疗后与对照组（P均〈0．001），治疗后的MMP-9水平与对照组比较差异无统计学意义（P=0．120）；病例组与对照组之间MMP-9基因R279Q、R668Q位点的RR、RQ、QQ基因型及R、Q等位基因频率分布比较差异无统计学意义（P均〉o．05）；R279Q、R668Q位点不同基因型病例组之间和对照组之间的血清MMP-9水平比较差异无统计学意义（P均〉0．05）。结论MMP-9与CAP的炎症反应有关，广东汉族人群MMP-9基因R279Q、R668Q位点多态性与CAP的遗传易感性和MMP-9的表达水平均无关。Objective To explore the association of matrix metalloproteinase - 9 （ MMP - 9 ） gene R279Q, R668Q polymorphisms with hereditary susceptibility of community -acquired pneumonia （CAP） and expression level of MMP - 9 in the population of Guangdong Han nationality. Methods 100 patients with CAP as the case group and 96 healthy subjects as the control group brought into study were all from the population of Guangdong Han nationality. The MMP - 9 serum level was tested by enzyme- linked immunosorbent assay （ELISA）. The MMP- 9 gene R279Q, R668Q polymorphisms of all subjects were detected by polymerase chain reaction - restriction fragment length polymorphism （PCR - RFLP）. Results The serum level of MMP - 9 of the case group in period of disease was significantly higher than that of the case group after treatment （ P 〈 0. 001 ）, as well as the control group （P〈0. 001 ）, and there was no significant difference between the case group after treatment and the control group（P =0. 120）. The distribution of RR, RQ, QQ genotypes and R, Q alleles of MMP -9 gene R279Q, R668Q between the two groups was not significant difference （all P 〉 0. 05 ）. There were no significant difference of MMP -9 serum level in different genotypes both in the case group and in the control group （ all P 〉 0.05 ）. Conclusion MMP - 9 relates to the inflammatory response of CAP. MMP -9 gene R279Q, R668Q polymorphisms arent associated with hereditary susceptibility of CAP and expression level in the population of Guangdong Han nationality.
