新生隐球菌临床分离株热休克反应的差别及其临床意义

摘　　要：目的：检测新生隐球菌中的热休克蛋白70（Hsp70）在不同的温度条件下表达量的差别，以探讨Hsp70在新生隐球菌致病中的作用。方法：采用鼠抗人、猪、狗等的Hsp70抗体通过Western印迹法检测6株临床分离株及1株CZYD2标准株新生隐球菌的 Hsp70，根据实验信号强弱定性判断在不同刺激条件下的 Hsp70产量。结果：在42℃×30min热应激及37℃×48h持续体外培养下新生隐球菌Hsp70产量较30℃培养下为高。结论：结合较难治愈病例分离株的Hsp70表达量高、热休克反应强的情况，推测Hsp70可能是新生隐球菌致病的一个因素。Objective: To study the role of Hsp70 in pathogensis in CryPtococcus neofOrmans under different tbermal cir-cumstances. Methods: Using the mouse's antibody to Hsp70 of human, pig, dog, etc., we detected 6 clinically isolatedstrains and 1 CZYD2 standard strain of Cryptococcus neoformans by Western blotting. According to the different hybridiza-tional signal, the production of Hsp70 was qualitatively determined. Results: The production of Hsp70 in the yeasts stressedby 42℃×30 min and cultured in 37℃×48 h was higher than that cu1tured in 30℃. Conclusion: The production of Hsp70 ismore in refractory cases, indicating patients infected by the yeasts with stronger heat shock reaction are more difficult to becured. So HSR has effects on pathogenesis in CryPtococcus neoformans I And the antibody of Hsp70 of the yeast may responsecrossing reaction with the Hsp70 of human. It may be a factor in pathogenesis of the yeast.
